摘要
目的:研究金匮肾气丸对肾小球肾炎大鼠的免疫调节作用及其可能机制。方法:采用尾静脉注射多柔比星建立肾小球肾炎大鼠模型,并将造模成功的30只大鼠随机分为3组:模型组、金匮肾气丸高(2.16 g·kg-1)、低(1.08 g·kg-1)剂量组,每组10只同时以10只未造模大鼠组成正常对照组。造模后,金匮肾气丸各剂量组分别灌胃给与相应剂量药物,正常对照组和模型组灌胃等体积的生理盐水。末次给药后,检测24 h尿蛋白、血清尿素氮、肌酐、肿瘤坏死因子-α(TNF-α)、白细胞介素6(IL-6)、C反应蛋白(CRP)等炎症因子及CD4+、CD8+等T淋巴细胞的表达水平用免疫组化法检测脾脏组织中CD4+、CD8+细胞蛋白表达水平。结果:与正常对照组比较,模型组大鼠24 h尿蛋白、血清尿素氮、肌酐、TNF-α、IL-6、CRP水平和CD8+细胞数量、CD8+/CD4+比值、CD8+细胞蛋白表达显著升高,CD4+细胞数量和蛋白表达显著降低(P<0.05);与模型组比较金匮肾气丸各剂量组大鼠24 h尿蛋白、血清尿素氮、肌酐、TNF-α、IL-6、CRP水平和CD8+细胞数量、CD8+/CD4+比值、CD8+细胞蛋白表达显著降低,CD4+细胞数量和CD4+细胞蛋白表达显著升高(P<0.05)。结论:金匮肾气丸对肾小球肾炎具有治疗作用,其机制可能与调节炎症细胞因子水平、调节脾脏CD4+、CD8+细胞表达有关。
Objective:To study the immunoregulatory effect of Jinkui Shenqi pills in rats with glomerulonephritis and its possible mechanism.Methods:Rat’s model of glomerulonephritis was established by injecting adriamycin into tail vein.Totally 30 rats with glomerulonephritis were randomly divided into 3 groups:the model group,Jinkui Shenqi pills high(2.16 g·kg-1)and low(1.08 g·kg-1)dose groups with 10 rats in each group,and another 10 rats without modeling were used as the normal control group.After the model establishment,Jinkui Shenqi pills groups were given corresponding drugs dose by intragastric administration,and the normal control group and the model group were given physiological saline with equal volume by intragastric administration.After the last administration,24 h urine of rats in each group was collected,and abdominal aorta blood was collected to detect 24 h urinary protein,serum urea nitrogen,creatinine,TNF-α,IL-6,CRP,CD4+and CD8+T lymphocyte expression levels,and immunohistochemistry was used to detect the expressions of CD4+and CD8+cells in the spleen of rats.Results:Compared with those in the normal control group,the24 h urinary protein,serum urea nitrogen,creatinine,TNF-α,IL-6,CRP levels,CD8+cell number,ratio of CD8+/CD4+and expression of CD8+cells in spleen tissue in the model group increased significantly,and the number and protein expression level of CD4+cells decreased significantly(P<0.05).Compared with those in the model group,the 24 h urinary protein,serum urea nitrogen,creatinine,TNF-α,IL-6,CRP levels,CD8+cell number,ratio of CD8+/CD4+and expression of CD8+cells in spleen tissue in Jinkui Shenqi pill groups decreased significantly,and the number and protein expression level of CD4+cells increased significantly(P<0.05).Conclusion:Jinkui Shenqi pills have therapeutic effect on glomerulonephritis.Its mechanism may be related to regulating inflammatory cytokines,and regulating the expressions of CD4+and CD8+cells in spleen.
