3味山姜属中药挥发油对胃溃疡寒证大鼠的药效机制研究 被引量:6

Study on Pharmacodynamic Mechanism of Volatile Oil from Three Chinese Medicines of Alpinia on Rat Model of Gastric Ulcer with Cold Syndrome

摘要 目的研究高良姜、红豆蔻、大高良姜3味山姜属中药挥发油对胃溃疡寒证大鼠的药效作用及其机制。方法通过灌胃冰知母溶液和15%乙酸建立大鼠胃溃疡寒证模型。空白对照组和模型组给予0.5%的吐温-80水溶液,阳性组给予干姜姜辣素溶液,受试药为高良姜、红豆蔻、大高良姜挥发油,分高、低剂量组分别给药,每天给药2次,共给药2d。检测大鼠造模后、末次给药后的大鼠体质量、肛温、进食量和饮水量;ELISA法检测胃组织胃泌素(Gastrin,GAS)、生长抑素(Somatostatin,SS)、6酮前列腺素F1α(6-keto-prostaglandin F1α,6-keto-PGF1α)及血清中皮质醇(Cortisol,COR)、胃动素(Motilin,MTL)、血栓素B2(Thromboxane-B2,TXB2)的水平。结果3味山姜属中药挥发油高剂量组大鼠饮食量、体质量及肛温均显著上升(P<0.01或P<0.05),大鼠GAS、SS、6-keto-PGF1α和血清中COR、MTL、TXB2水平显著降低(P<0.01或P<0.05),SS、6-keto-PGF1α水平显著升高(P<0.01)。3味山姜属中药挥发油低剂量组大鼠的肛温、SS、6-keto-PGF1α水平均升高、MTL水平显著降低(P<0.01或P<0.05)。红豆蔻挥发油低剂量组大鼠进食量及饮水量升高、COR水平降低(P<0.01或P<0.05)。大高良姜挥发油低剂量组大鼠饮水量升高(P<0.01)。结论高良姜、红豆蔻、大高良姜的挥发油对胃溃疡寒证大鼠具有治疗作用,其药效机制为通过降低大鼠GAS、MTL、COR、TXB2水平,升高SS、6-keto-PGF1α水平,以调节胃肠激素及神经递质的平衡,改善胃黏膜微循环,增加局部血流量,减少血栓形成,发挥保护胃黏膜损伤及促进胃溃疡愈合的作用。 Objective To study the pharmacodynamic effect and mechanism of volatile oil from Gaoliangjiang(Alpinia officinarum Hance),Hongdoukou(Alpinia galanga Willd)and Dagaoliangjiang(Alpinia galanga Willd)on rats with gastric ulcer cold syndrome.Methods The rat model of gastric ulcer with cold syndrome was established by intragastric administration of cold solution of Zhimu(Rhizoma Anemarrhenae)and 15%glacial acetic acid.After modeling,the blank control group and model group were given 0.5%Tween-80 aqueous solution,the gingerol group was given gingerol solution,and the high-dose and low-dose groups of Gaoliangjiang(Alpinia officinarum Hance),Hongdoukou(Alpinia galanga Willd)and Dagaoliangjiang(Alpinia galanga Willd)volatile oil were given the volatile oil solution of each drug respectively twice a day for 2 days.The body weight,anal temperature,food intake and water intake of rats after modeling and the last administration were recorded.The levels of GAS,SS,6-keto-PGF1αin gastric tissue and COR,MTL and TXB2 in serum were detected by ELISA.Results After treatment,the diet,body weight and rectal temperature of rats in the high-dose group of three Chinese medicines of Alpinia volatile oil were increased significantly(P<0.01 or P<0.05).The contents of GAS,MTL,COR and TXB2 in rats were significantly reduced(P<0.01 or P<0.05),and the contents of SS and 6-keto-PGF1αwere significantly increased(P<0.01).The rectal temperature and contents of SS and 6-keto-PGF1αof rats in the low-dose group of volatile oil of three Chinese medicines of Alpinia were increased,and the content of MTL was significantly decreased(P<0.01 or P<0.05).In the low-dose of Hongdoukou(Alpinia galanga Willd)volatile oil group,the food intake and water intake were increased,and the COR content was decreased(P<0.01 or P<0.05).The water consumption of rats in the low-dose group of Hongdoukou(Alpinia galanga Willd)volatile oil was increased(P<0.01).Conclusion The volatile oils of Gaoliangjiang(Alpinia officinarum Hance),Hongdoukou(Alpinia galanga Willd)and Dagaoliangjiang(Alpinia galanga Willd)have a certain therapeutic effect on rats with gastric ulcer cold syndrome.The mechanism may be through reducing the contents of GAS,MTL,COR and TXB2 and increasing contents of SS and 6-keto-PGF1αto adjust the balance of gastrointestinal hormones and neurotransmitters,improve gastric mucosal microcirculation,increase local blood flow,reduce thrombosis and play a role in protecting gastric mucosal damage and promoting gastric ulcer healing.
