摘要
目的:为提升南药巴戟天现代种业竞争力,应用生物技术优化组培苗驯化移栽过程,建立高效、优质的种苗繁育技术体系。方法:以肇庆道地巴戟天脱毒组培苗为材料,研究不同基质配比和生根时长对其驯化移栽的成效。结果:河沙和黄泥基质的有机质及有效矿质元素含量较低,不适宜巴戟天组培苗的生长。组培苗在泥炭土与河沙的组合基质中成活率较高,150 d时成活率保持在70.03%~75.33%;其中组合基质(泥炭土∶河沙=2∶1,V/V)最有利于组培苗的驯化移栽,种植150 d时,幼苗的根系长度、株高、叶长及地上部鲜重均显著高于其他基质中的幼苗。生根时长为2月龄的巴戟天组培苗,移栽的成活率、株高和叶片的生长状况优于4月龄和8月龄的组培苗。结论:在气候大棚中,取2月龄巴戟天组培苗,放置于大棚驯化7~10 d后,移栽至组合基质(泥炭土∶河沙=2∶1,V/V)中驯化培养120~150 d,株高可达15 cm左右,节数8~9个,对生叶数约14片,生长的种苗整齐、质量好,完成组培苗向种苗的过渡,适合规模化生产。
Objective: To optimize the acclimation and transplanting process of tissue culture seedlings by biotechnology, and to establish an efficient and high-quality breeding technology system of seedlings, so as to enhance the competitiveness of modern seed industry of Morinda officinalis.Methods: Using the detoxified tissue culture seedlings of Morinda officinalis in Zhaoqing as the material, the effects of different medium ratio and rooting duration on the acclimation and transplanting were studied.Results: The contents of organic matter and available mineral elements in river sand and yellow mud substrates were low, which was not suitable for the growth of Morinda officinalis tissue culture seedlings.The survival rate of Morinda officinalis tissue culture seedlings in peat soil and river sand combined substrate was higher, and the survival rate was 70.03%to 75.33%at 150 days.The combined matrix(peat soil∶river sand=2∶1,V/V) was most conducive to the issue culture seedlings on the acclimation and transplanting, After 150 days of planting, the root length, plant height, leaf length and fresh weight of aerial part of seedlings were significantly higher than those of seedlings in other substrates.The survival rate, plant height and leaf growth condition of two-month-old tissue culture seedlings were better than those of four-month-old and eight-month-old tissue culture seedlings.Conclusion: In the greenhouse climate, the two-month-old seedlings are cultured in the greenhouse for 7 to 10 days, and then transplanted to the combined matrix(peat soil∶river sand=2∶1,V/V) for 120 to 150 days.The plant height is about 15 cm, the number of nodes is 8 to 9,and the number of opposite leaves is about 14.The transition from tissue culture seedlings to production seedlings is well completed and the seedlings is tidy, superior in quality, which is suitable for large-scale production.
