摘要 目的:应用超高效液相色谱-四极杆/静电场轨道阱高分辨质谱(UPLC-Q-Orbitrap HRMS)技术,对真武汤中的化学成分进行定性分析。方法:色谱柱为Waters ACQUITY UPLC^(■)HSS C_(18)(150 mm×2.1 mm,1.8μm);以乙腈-0.1%甲酸水溶液为流动相,梯度洗脱;体积流量0.2 mL/min;进样量5μL;柱温30℃。采用电喷雾离子源,正、负离子扫描模式下采集质谱信息,运用Compound Discoverer 3.0软件将未知化合物的一级、二级质谱信息与对照品、参考文献及数据库中的化合物质谱信息进行比对分析。结果:从真武汤中共指认出120个化合物,包括生物碱类、内酯类、单萜苷类、三萜酸类、姜辣素类、有机酸类、倍半萜类、香豆素类、醛类及苷类化合物。其中22个化合物经对照品验证。结论:该方法可快速、准确分析真武汤的化学成分,为真武汤药效物质基础研究提供实验依据。 Objective:To qualitatively analyze the chemical constituents of Zhenwu decoction by UPLC-Q-Orbitrap HRMS.Methods:The determination was performed on a Waters ACQUITY UPLC^(■)HSS C_(18)(2.1 mm×150 mm,1.8μm)chromatographic column,mobile phase was acetonitrile-0.1%formic acid solution with gradient elution,flow rate was 0.2 mL/min,injection volume was 5μL,column temperature was 30℃.The mass spectrum information was collected in positive and negative ion scanning mode by electron spray ionization.Compound Discoverer 3.0 software was used to compare and analyze the primary and secondary mass spectrum information of the target compounds with the fragment ion peaks and fragmentation patterns of the compound in reference substances,references and databases.Results:A total of 120 compounds in Zhenwu decoction were identified,including alkaloids,lactones,monoterpene glycosides,triterpenoid acids,gingerols,organic acids,sesquiterpenoids,coumarins,aldehydes and glycoside,among which 22 compounds were verified by reference substances.Conclusion:This method can quickly and accurately analyze the chemical components of Zhenwu decoction,and provide experimental basis for the pharmacodynamic substances study of Zhenwu decoction.
