摘要
目的:探究黑眶蟾蜍的遗传多样性水平和遗传结构,以期为蟾酥基原鉴定、种质评价以及资源保护和利用等方面提供理论依据。方法:采用高通量测序技术,对黑眶蟾蜍9个地理种群共54个体进行简化基因组测序,挖掘其单核苷酸多态性位点,分析其遗传多样性和遗传结构。结果:基于获得的10724个高质量单核苷酸多态性位点分析发现,黑眶蟾蜍的9个地理种群的期望杂合度值(He)为0.2167~0.5870,观测杂合度值(Ho)为0.2803~0.7118,核苷酸多样性为0.1461~0.1825。分子方差分析表明,种群遗传差异主要来源于个体间,且差异达显著水平(P<0.05)。系统进化树、主成分分析与种群遗传结构分析均将样品聚成2个不同的遗传类群。结论:该研究在基因组水平确定了黑眶蟾蜍的遗传分化关系,为今后优质品种选育以及蟾酥道地性形成的分子机制积累了研究基础。
Objective:To explore the level of genetic diversity and genetic structure of Bufo melanostictus,so as to provide theoretical basis for the primitive identification,germplasm evaluation and resource protection and utilization of Bufonis Venenum.Methods:A total of 54 individuals from 9 geographic populations of Bufo melanostictus were sequenced by high-throughput sequencing technology,and single nucleotide polymorphisms(SNPS)were extracted to analyze their genetic diversity and structure.Results:Based on the obtained 10724 high quality SINGLE nucleotide polymorphisms(SNPS),the expected heterozygosity(He),observed heterozygosity(Ho)and nucleotide diversity of Bufo melanostictus from 9 geographic populations ranged from 0.2167 to 0.5870,0.2803 to 0.7118,0.1461 to 0.1825.Molecular variance analysis showed that the genetic difference of population was mainly from individuals,and the difference was significant(P<0.05).Phylogenetic tree,principal component analysis and population genetic structure analysis all clustered the samples into two different genetic groups.Conclusion:This study confirms the genetic differentiation relationship of Bufo melanostictus at the genome level,and provides a basis for the future breeding of high-quality varieties and the molecular mechanism of the formation of Bufonis Venenum quality.
