黄金艾蒿的非靶向代谢组和转录组学分析 被引量:7

摘要 目的:鉴定黄金艾蒿中次生代谢物质及其合成的关键基因。方法:通过非靶向代谢组学鉴定黄金艾蒿叶片中的次生代谢物质成分,利用转录组测序分析主要代谢成分合成途径的关键基因。结果:基于非靶向代谢组在黄金艾蒿叶片中鉴定出氨基酸类、生物碱类、奎宁酸及其衍生物类以及黄酮类等共98种代谢物,其中黄酮类物质有21种,包括圣草素、柚皮素、异鼠李素、异牡荆素以及芹菜素和山柰酚的糖苷衍生物等。基于转录组测序,经过de novo组装、去冗余后共获得45494条Unigene,平均长度为702.17 bp,N50为830 bp。经过与公共数据库进行同源比对,共有36866(81.03%)条Unigene获得功能注释,其中15025(33.03%)条Unigene在KEGG数据库中得到注释,共涉及129个代谢通路。根据基因功能进一步鉴定到53个Unigene参与类黄酮的生物合成,包括黄酮类合成的关键酶PAL、CHS、CHI、F3H和FLS等。同时还鉴定到245个细胞色素P450家族基因和74个糖基转移酶家族基因。结论:该研究结果为下一步有针对性地研究黄金艾蒿的代谢物质活性和开展黄酮类化合物合成关键基因的功能研究奠定了基础。 Objective:To identify the key genes of secondary metabolites and their synthesis in Artemisia vulgaris‘Variegate’.Methods:The secondary metabolites in the leaves of Artemisia vulgaris‘Variegate’were identified by non-targeted metabolomics.Transcriptome sequencing was used to analyze the key genes in the main metabolic component synthesis pathway.Results:Ninety-eight metabolites,including amino acids,alkaloids,quinine acid and its derivatives and flavonoids,were identified in the leaves of Artemisia vulgaris‘Variegate’on the basis of non-targeted metabolomics.Among them,there were 21 kinds of flavonoids,including eriodictyol,naringenin,isorhamnetin,isovitexin,apigenin and kaempferol glycoside derivatives.Based on transcriptome sequencing,a total of 45494 Unigenes were obtained through de novo assembly after redundancy removal,with an average length of 702.17 bp and an N50 of 830 bp.After homologous comparison with the public database,a total of 36866(81.03%)Unigenes obtained functional comments,among them,15025(33.03%)Unigenes were annotated in KEGG database and involved in 129 metabolic pathways.According to the gene function,53 Unigenes were further identified to participate in the biosynthesis of flavonoids,including the key enzymes involved in the synthesis of flavonoids PAL,CHS,CHI,F3 H and FLS.At the same time,245 cytochrome P450 family genes and 74 glycosyltransferase family genes were identified.Conclusion:The results of this study provide a basis for further research on the metabolic activity and the function of key genes in the synthesis of flavonoids of Artemisia vulgaris‘Variegate’.
