结直肠癌患者EBV感染与癌组织COX-2、P-gp、p53蛋白表达相关性 被引量:5

Correlation between EB virus infection and expression levels of COX-2, P-glycoprotein and p53 protein in cancer tissues of patients with colorectal cancer

摘要 目的探讨结直肠癌患者爱泼斯坦-巴尔病毒(EBV)感染与癌组织中环氧化酶-2(COX-2)、P-糖蛋白(P-gp)、p53蛋白表达的相关性。方法选取2018年1月-2019年12月间海南省人民医院收治的114例结直肠癌手术患者,采用原位分子杂交检测EB病毒编码的小RNA(EBER)表达,分析结直肠癌患者EBV感染与临床病理、COX-2、P-gp、p53蛋白表达的关系,分析EBER表达与结直肠癌临床病理特征的关系,及与癌组织COX-2、P-gp、p53蛋白表达水平的关系。结果癌组织EBER阳性率高于癌旁组织(P<0.05);结直肠癌EBER阳性临床分期、肿瘤分化程度和淋巴结转移情况与EBER阴性者比较,差异有统计学意义(P<0.05);结直肠癌EBER阳性者COX-2、P-gp、p53蛋白阳性率均高于EBER阴性者(P<0.05);结直肠癌癌组织COX-2、P-gp、p53蛋白阳性表达与EBER阳性表达均呈正相关关系(P<0.05);P-gp阳性是预测结直肠癌EBV感染的有效指标(P<0.05);P-gp是影响结直肠癌合并EBV感染的独立危险因素(OR=2.399,P=0.018,95%CI:1.164~4.944)。结论EBV感染与结直肠癌患者COX-2、P-gp、p53高表达相关,COX-2、P-gp、p53高表达可能是EBV相关结直肠癌患者病情进展的基础。 OBJECTIVE To explore the correlation between Epstein-Barr virus(EBV)infection and expression levels of cyclooxygenase-2(COX-2),P-glycoprotein(P-gp)and p53 protein in cancer tissues in patients with colorectal cancer.METHODS Specimens of cancer tissue and adjacent tissue of 114 patients undergoing colorectal cancer surgery in Hainan General Hospital between Jan 2018 and Dec 2019 were recruited.The expression of EB virus-encoded small RNA(EBER)was detected by in situ hybridization,and the relationship between EBV infection and clinicopathology and expressions of COX-2,P-GP and p53 protein was analyzed among patients with colorectal cancer.The relationship between EBER expression and clinicopathological characteristics of colorectal cancer,and the expression levels of COX-2,P-gp and p53 protein in cancer tissues were analyzed.RESULTS The positive rate of EBER of cancer tissues was significantly higher than that of adjacent tissues(P<0.05).There were significant differences in the clinical stage,tumor differentiation and lymph node metastasis between EBER positive patients with colorectal cancer and EBER negative patients(P<0.05).The positive rates of COX-2,P-gp and p53 protein in EBER positive patients with colorectal cancer were significantly higher than those in EBER negative patients(P<0.05).The positive expression of COX-2,P-gp and p53 protein in colorectal cancer tissues were positively correlated with positive expression of EBER(P<0.05).Positive P-gp was an effective index for predicting EBV infection in colorectal cancer(P<0.05).P-gp was an independent risk factor for colorectal cancer with EBV infection(OR=2.399,P=0.018,95%CI:1.164~4.944).CONCLUSION EBV infection is associated with the high expression levels of COX-2,P-gp and p53 among patients with colorectal cancer.The high expression of COX-2,P-gp and p53 may be the basic indexes for the progression of patients with EBV-related colorectal cancer.
