猴头菌-青蒿生物转化物中1个新的倍半萜内酯成分 被引量:1

A new sesquiterpene lactone from biological transformation of Hericium erinaceus and Artemisiae Annuae Herba

摘要 研究猴头菌-青蒿生物转化物(HQ生物转化物)的化学成分。综合运用硅胶柱色谱、Sephadex LH-20凝胶柱色谱和半制备高效液相色谱等分离技术,对HQ生物转化物75%乙醇提取物乙酸乙酯萃取部位进行系统分离纯化,通过化合物理化性质、波谱数据以及与文献数据比对等进行结构鉴定。从中共分离鉴定得到9个化合物:2α-羟基脱氧青蒿素(1)、6β-羟基-豆甾-4,22-二烯-3-酮(2)、3β,5α-二羟基麦角甾-7,22-二烯-6-酮(3)、木栓酮(4)、dankasterone(5)、ergosterol endoperoxide(6)、3β-羟基-5,9-环氧-(22E,24R)-麦角甾-7,22-二烯-6-酮(7)、3β,5α,9α-三羟基-(22E,24R)-麦角甾-7,22-二烯-6-酮(8)、豆甾烷-3-酮(9)。其中化合物1为新的倍半萜内酯类成分,命名为2α-羟基脱氧青蒿素。运用琼脂扩散法和药敏纸片法测定了化合物1~9抗幽门螺旋杆菌(Hp)的活性,结果显示化合物1、2、5具有一定的抗Hp活性。 This study aimed to investigate the chemical constituents from the biological transformation of Hericium erinaceus and Artemisiae Annuae Herba(HQ biological transformation). The chemical constituents of ethyl acetate fraction of 75% ethanol extract in HQ biological transformation were separated and purified by silica gel and Sephadex LH-20 gel column chromatographies together with semi-preparative high performance liquid chromatography(HPLC). Their structures were identified by physicochemical properties, spectroscopic analysis, as well as comparisons with the data reported in literature. Nine compounds were isolated and identified as 2α-hydroxydeoxyartemisinin(1), 6β-hydroxy-stigmast-4,22-dien-3-one(2), 3β,5α-dihydroxy-ergosta-7,22-dien-6-one(3), friedelin(4), dankasterone(5), ergosterol endoperoxide(6), 3β-hydroxy-5,9-epoxy-(22E,24R)-ergosta-7,22-dien-6-one(7), 3α,5α,9α-trihydroxy-(22E,24R)-ergosta-7,22-dien-6-one(8), and stigmast-3-one(9). Compound 1 was a new sesquiterpene lactone named 2α-hydroxy-deoxyartemisinin. The activity against Helicobacter pylori(Hp) of compounds 1-9 in vitro was determined by Kirby-Bauer disk diffusion method. The screening results showed compounds 1, 2 and 5 had certain anti-Hp activity.
