摘要
建立一种固相萃取-高效液相色谱法测定食品中8种合成着色剂的方法。样品经提取液超声提取,离心后上清液经Pro Elut PWA-2固相萃取柱净化。以甲醇和0.02 mol/L乙酸铵水溶液为流动相,用Thermo Acclaim^TM 120 C18色谱柱进行梯度洗脱分离,DAD检测器多波长检测,外标法定量测定。结果表明,8种合成着色剂在1~50μg/mL质量浓度范围内线性关系良好,相关系数均大于0.999,方法检出限为0.01~0.03 mg/kg,定量限为0.03~0.10 mg/kg。在加标量0.5,5.0和10.0 mg/kg水平下,样品加标平均回收率为86.5%~99.5%,相对标准偏差为0.5%~2.7%(n=6)。该方法灵敏度高、重现性好、分析速度快,适用于食品中8种合成着色剂的同时检测。
A high performance liquid chromatography with solid phase extraction method for analysis of 8 kinds of synthetic colorants in food was established.The samples were extracted ultrasonically with extraction agent.After centrifugation,the supernatant was purified by ProElut PWA-2 column.Methanol and 0.02 mol/L aletic acid ammonium were selected as gradient,the analysis was performed on Thermo Acclaim^TM 120 C18 column,DAD detector was used to test samples by multiple wavelengths,and external standard method was used for quantitative analysis.The results showed a good linear correlation in the concentration between 1μg/mL and 50μg/mL.The correlation coefficient was greater than 0.999.The detection limit of them was 0.01-0.03 mg/kg.The limit of quantitation was 0.03-0.10 mg/kg.The average recoveries of 8 kinds of synthetic colorants were 86.5%-99.5%,and the RSDs were 0.5%-2.7%(n=6)at 3 levels,such as 0.5,5.0 and 10.0 mg/kg.The method was proved to be sensitive,reproducible,quick and adapts to the simultaneous determination of 8 kinds of synthetic colorants in food.
