摘要
目的:优选人参天麻颗粒的最佳水提工艺。方法:建立高效液相色谱紫外检测器测定人参天麻颗粒中天麻素含量的测定方法;以天麻素提取率结合干膏得率为考察指标,分别选择加水量(A)、煎煮时间(B)、煎煮次数(C)为考察因素,以L_(9)(3^(4))进行正交试验,用双指标综合评分法,依据实验结果优选最佳水提取工艺。结果:本次实验建立了人参天麻颗粒中天麻素含量的测定方法;以天麻素提取率为评价指标,最佳条件及影响因素C3>B3>A3;以干膏得率为评价指标,最佳条件及影响因素C3>B2>A3。加权评分优化,又考虑生产实际,最终确定最佳水提条件为A_(2)B_(2)C_(2),即加8倍量的水,提取二次,每次提取2 h。结论:天麻素含量测定方法灵敏可靠,重现性好,优选的水提取工艺稳定可行,能用于人参天麻颗粒的规模化生产。
Objective:To establish the best water extraction process of Renshentianma Granules.Methods:To establish content determination way of Gastrodin in Renshentianma Granules by HPLC UV detector.Take extraction rate of Gastrodin combined with yield of dry extract as the indecator,separate selection factor of amount of water added(A),boiling time(B)and number of decoctions(C),where the L_(9)(3^(4))orthogonal test was carried out.Selection two-index comprehensive scoring method,the best water extraction process was established by result of orthogonal Test.Results:to establish content determination of Gastrodin in Renshentianma Granules,the extraction rate of Gastrodin were set as evaluation indexes.The optimum condition and Influencing factor were C3>B3>A3.Set yield of dry extract as evaluation indexes,where optimum condition and Influencing factor were C3>B2>A3.When scoring optimization and actual production,the best condition was A_(2)B_(2)C_(2),where adding 8 times amount of water,extractiong twice,boiling 2 hours each time.Conclusion:The method is sensitive,reliable and reproducible,and the optimized water extraction process is stable and feasible,which can be used for the large-scale production of Renshentianma Granules.
