HBV感染与原发性肝细胞癌组织MAPK信号通路蛋白表达水平的关联性 被引量:3

Correlation between HBV infection and the expressions of MAPK signaling pathway proteins in primary hepatocellular carcinoma tissues

摘要 目的探究慢性乙型肝炎病毒(HBV)感染与原发性肝细胞癌(PHC)病理特征及癌组织丝裂酶原活化蛋白激酶(MAPK)信号通路蛋白表达的关联性。方法以2016年1月-2021年2月PHC患者78例为研究对象,根据HBV感染情况分为HBV感染组(n=41)及未感染组(n=37),比较不同Edmondson分级患者的HBV DNA载量及乙型肝炎表面抗原(HBsAg)、乙型肝炎病毒e抗原(HBeAg)定量;比较HBV感染组及未感染组癌组织、癌旁组织MAPK信号通路蛋白表达情况,分析HBV感染与病理特征及癌组织MAPK信号通路蛋白表达情况的关系,并分析PHC患者HBV DNA载量与癌组织MAPK信号通路蛋白表达情况的相关性。结果Edmondson分级Ⅲ级患者的HBV DNA载量及HBsAg、HBeAg定量均高于Ⅱ级患者,差异有统计学意义(t=3.022、3.319、4.934,P均<0.05);HBV DNA载量及HBsAg、HBeAg定量与Edmondson分级成正相关(r=0.440、0.289、0.430,P均<0.05);感染组及未感染组癌组织细胞外信号调节激酶(ERK)1、ERK2、p38的IOD值均大于癌旁组织,且感染组癌组织ERK1、ERK2的IOD值大于未感染组癌组织,差异有统计学意义(P<0.05);感染组及未感染组癌组织c-Jun N端激酶1(JNK1)的IOD值小于癌旁组织,差异有统计学意义(P<0.05);HBV感染是导致PHC癌组织ERK1蛋白表达量升高的危险因素(P<0.05);HBV感染的PHC患者HBV DNA载量与癌组织ERK1、ERK2、p38蛋白表达成正相关(r=0.580、0.609、0.439,P均<0.05)。结论PHC患者HBV DNA载量及HBsAg、HBeAg定量与Edmondson分级相关,且患者HBV DNA载量与癌组织ERK1、ERK2、p38蛋白表达水平相关。 Objective To explore the correlations between hepatitis B virus(HBV)infection and pathological characteristicsof primary hepatocellular carcinoma(PHC),expressions of mitogen-activated protein kinase(MAPK)signaling pathwayproteins in carcinoma tissues.MethodsA total of 78 patients with PHC were enrolled as the research objects betweenJanuary 2016 and February 2021.According to presence or absence of HBV infection,they were divided into infection group(n=41)and non-infection group(n=37).HBV DNA load,hepatitis B surface antigen(HBs Ag)and hepatitis B e antigen(HBe Ag)quantification in patients with different Edmondson grading were compared.The expressions of MAPK signalingpathway proteins in carcinoma tissues and para-carcinoma tissues were compared between infection group and non-infectiongroup.The relationship between HBV infection and pathological characteristics,expressions of MAPK signaling pathwayproteins in carcinoma tissues,and the correlation between HBV DNA load and expressions of MAPK signaling pathwayproteins in carcinoma tissues were analyzed.ResultsHBV DNA load,HBs Ag and HBe Ag quantification in patients withEdmondson grading at gradeⅢwere higher than those at gradeⅡ(t=3.022,3.319,4.934,all P<0.05).HBV DNA load,HBs Ag and HBe Ag quantification were positively correlated with Edmondson grading(r=0.440,0.289,0.430,all P<0.05).Ininfection group and non-infection group,IOD values of extracellular signal-regulated kinase(ERK)1,ERK2 and p38 incarcinoma tissues were greater than those in para-carcinoma tissues.IOD values of ERK1 and ERK2 in carcinoma tissues ofinfection group were greater than those of non-infection group(P<0.05).In infection group and non-infection group,IOD value of c-Jun NH2-terminal kinase1(JNK1)in carcinoma tissues was lower than that in para-carcinoma tissues(P<0.05).HBV infection was a risk factor causing the increase of ERK1 protein in PHC tissues(P<0.05).HBV DNA load was positivelycorrelated with expressions of ERK1,ERK2 and p38 proteins in carcinoma tissues of PHC patients with HBV infection(r=0.580,0.609,0.439,P<0.05).ConclusionsHBV DNA load,HBs Ag and HBe Ag quantification were correlated withEdmondson grading in PHC patients.HBV DNA load was related to the expressions of ERK1,ERK2 and p38 proteins incarcinoma tissues.
