摘要
目的探讨重度子痫前期孕妇胎盘绒毛膜板动脉(CPAs)血管中微小RNA-217(miR-217)的表达情况及与脂代谢的关系。方法选取西南医科大学附属医院2020年10月-2021年11月产科住院患者,采用实时荧光定量PCR法分别测量miR-217在正常孕妇(NP)组(20例)、晚发型重度子痫前期(LSPE)组(20例)、早发型重度子痫前期(ESPE)组(20例)CPAs血管中的表达量;比较三组孕妇终止妊娠前的脂代谢水平,并分析CPAs血管中miR-217表达量与脂代谢水平间的关系。结果NP、LSPE、ESPE三组孕妇的CPAs血管中miR-217的ΔCt值分别为(9.76±0.74)、(8.25±0.98)、(6.90±0.59),三组间差异有统计学意义(F=66.626,P<0.05);ESPE、LSPE组的总胆固醇(TC)、甘油三酯(TG)、低密度脂蛋白胆固醇(LDL-C)水平高于NP组(F=3.872、8.563、4.907),ESPE、LSPE组的高密度脂蛋白胆固醇(HDL-C)水平低于NP组(F=5.196),差异均有统计学意义(P<0.05);多元线性回归分析显示CPAs血管中的miR-217含量与TC、TG、LDL-C水平成正相关,与HDL-C水平成负相关。结论miR-217可能通过诱导脂代谢紊乱参与重度子痫前期的发病,且miR-217表达量可能与重度子痫前期病情的严重程度有关,miR-217可能成为治疗子痫前期的另一个新方向。
Objective To investigate the expression of microRNA-217(miR-217)in the placental chorionic plate arteries(CPAs)in pregnant women with severe preeclampsia and its relationship with lipid metabolism.Methods Obstetrics inpatients were selected from October 2020 to November 2021.Real-time quantitative PCR was used to measure miR-217 in20 cases of normal pregnancy(NP),20 cases of late-onset severe preeclampsia(LSPE)and 20 cases of early-onset severe preeclampsia(ESPE).The lipid metabolism levels of pregnant women in the three groups before termination of pregnancy were compared,and the relationship between miR-217 expression level in CPAs vessels and lipid metabolism level was analyzed.Results TheΔCt values of miR-217 in CPAs vessels of NP,LSPE and ESPE groups were(9.76±0.74),(8.25±0.98)and(6.90±0.59),respectively,with statistical significance(F=66.626,P<0.05).Total cholesterol(TC),triglyceride(TG)and low density lipoprotein cholesterol(LDL-C)levels in ESPE and LSPE groups were higher than those in NP group(F=3.872,8.563,4.907).High-density lipoprotein cholesterol(HDL-C)levels in ESPE and LSPE groups were lower than those in NP group(F=5.196),the differences were statistically significant(P<0.05).Multiple linear regression analysis showed that the miR-217 content in CPAs vessels was positively correlated with TC,TG and LDL-C levels,and negatively correlated with HDL-C levels.Conclusions MiR-217 might be involved in the pathogenesis of severe preeclampsia by inducing lipid metabolism disorder,and the expression level of miR-217 might be related to the severity of severe preeclampsia.Therefore,miR-217 might become another new direction for the treatment of preeclampsia.
