摘要
目的探讨茯苓水提物对环磷酰胺荷瘤小鼠免疫微环境的影响。方法2020年5月将40只小鼠随机分为空白组、模型组和茯苓水提物低、中、高剂量组,各8只。空白组不予造模,其余各组均建立环磷酰胺荷瘤小鼠模型,从造模当日起,空白组和模型组均采用等体积0.9%氯化钠溶液灌胃;茯苓水提物低、中、高剂量组灌胃剂量分别为15 mg/kg、30 mg/kg、60 mg/kg;连续灌胃28 d。观察小鼠脾脏、胸腺指数、脾脏NK细胞及CD_(4)^(+)、CD_(8)^(+)细胞比例、NK细胞活性。结果与空白组比较,造模组小鼠胸腺指数降低、脾脏指数升高(P<0.05);与造模组比较,茯苓水提物低、中、高剂量组小鼠胸腺指数升高,脾脏指数降低(P<0.05)。与空白组比较,造模组小鼠脾脏NK细胞比例降低(P<0.01);与造模组比较,茯苓水提物低、中、高剂量组小鼠脾脏NK细胞比例均增加(P<0.05)。与空白组比较,造模组小鼠脾脏CD_(4)^(+)细胞比例降低、CD_(8)^(+)细胞比例升高(P<0.01);与造模组比较,茯苓水提物低、中、高剂量组小鼠脾脏CD_(4)^(+)细胞比例升高、CD_(8)^(+)细胞比例降低(P<0.05)。与空白组比较,造模组小鼠NK细胞活性降低(P<0.01);与造模组比较,茯苓水提物低、中、高剂量组小鼠NK细胞活性均增加(P<0.05)。结论茯苓水提物改善环磷酰胺引起的毒副作用,可能是通过增加脾脏NK、CD_(4)^(+)、CD_(8)^(+)细胞比例及增强NK细胞的活性发挥免疫调控作用。
Objective To investigate the effect of Poria cocos water extract on immune microenvironment of cyclophosphamide tumor bearing mice.Methods In May 2020,40 mice were randomly divided into blank group,model group and low,medium and high dose groups of Poria cocos water extract,with 8 mice in each group.No model was made in the blank group,and cyclophosphamide tumor bearing mouse models were established in other groups.From the day of modeling,the blank group and model group were perfused with equal volume of 0.9%sodium chloride solution;The intragastric doses of Poria cocos water extract in low,medium and high dose groups were 15 mg/kg,30 mg/kg and 60 mg/kg respectively;Continuous gavage for 28 days.The indexes of spleen and thymus,the proportion of spleen NK cells and CD_(4)^(+),CD_(8)^(+)cells,and the activity of NK cells were observed.Results Compared with the blank group,the thymus index decreased and the spleen index increased in the model group(P<0.05);Compared with the model group,the thymus index increased and the spleen index decreased in the low,medium and high dose groups of Poria cocos water extract(P<0.05).Compared with the blank group,the proportion of spleen NK cells in the model mice decreased(P<0.01);Compared with the model group,the proportion of spleen NK cells in low,medium and high dose groups of Poria cocos water extract increased(P<0.05).Compared with the blank group,the proportion of CD_(4)^(+) and CD_(8)^(+) cells in spleen of model mice decreased(P<0.01);Compared with the model group,the proportion of CD_(4)^(+) and CD_(8)^(+) cells in spleen of mice in low,medium and high dose groups of Poria cocos water extract increased(P<0.05).Compared with the blank group,the activity of NK cells in the model mice decreased(P<0.01);Compared with the model group,the activity of NK cells in low,medium and high dose groups of Poria cocos water extract increased(P<0.05).Conclusion Poria cocos water extract can improve the toxic and side effects caused by cyclophosphamide,which may play an immune regulatory role by increasing the proportion of spleen NK,CD_(4)^(+),CD_(8)^(+) cells and enhancing the activity of NK cells.
