地黄饮子对APP/PS1小鼠IL-1、TNF-α和NF-κB p65的影响 被引量:15

Effects of Dihuang Yinzion IL-1,TNF-α and NF-κB p65 in App/Ps1 Mice

摘要 目的研究地黄饮子(DHYZ)对APP/PS1双转基因痴呆小鼠炎症因子和核转录因子(NF-κB)的影响。方法将35只APP/PS1双转基因痴呆小鼠随机分为模型组、DHYZ组;另以同背景同月龄的C 57 BL/6 J阴性小鼠为空白组。DHYZ给予地黄饮子相当于生药9.75 g/(kg·d)灌胃,模型组和空白组给予等量蒸馏水灌胃。各组灌胃30天后进行Mirror水迷宫空间探索试验后取材。HE染色法观察皮层及海马CA1区神经细胞的形态结构;尼氏染色法观察神经元;免疫组化方法检测炎症因子TNF-α、IL-1β;Western印迹检测法检验磷酸化NF-κB p65(pp65)的表达。结果DHYZ降低APP/PS1小鼠皮层和海马组织的TNF-α、IL-1β(P<0.01)和皮层区NF-κB pp65(P<0.01),进而改善皮层及海马CA1区细胞的形态结构和增加皮层及海马区的尼氏小体数量。结论DHYZ改善学习记忆能力可能通过抑制pp65,下调细胞炎性因子IL-1和TNF-α的释放水平,使皮层及海马神经元损伤减少。 Objective This experiment is to investigate the effects of Dihuang Yinzi(DHYZ)on inflammatory factors and NF-κBp65 in APP/PS1 mice.Methods The APP/PS1 dementia models were randomly divided into model groups and Cchinese medicine groups.The blank groups had C 57 BL/6 J mice.The models in Chinese medicine group were given intragastric administration with DHYZ 9.75 g/(kg·d).The model group and blank group were treated with distilled water.After 30 days,we detected the abilities of learning and memory of mice by Morris water maze.And then,we observedthe Nissl’s stain and HE stain.The expressions of inflammatory factor in the hippocampus and cortex were detected by immunohistochemistry.We analyzed the expressions of phospho-NF-κBp65(pp65)through Western blot and immunofluorescence analysis in the cortex and hippocampus of APP/PS1 mice.Results Behavioral results showed that DHYZ significantly increased the distance of platform crossings on the fifth day(P<0.01).In DHYZ group,the lesions in cortex and CA1 regions of hippocampus were lighter than those in group APP/PS1 and some cell bodies became atrophic.DHYZ increased the number of Nissl bodies in cortex and hippocampus.DHYZ decreased the levels of inflammatory factors including IL-1βand TNF-α(P<0.01).DHYZ could depress the expressions of pp65 in the cortex of APP/PS1 mice(P<0.01).Conclusion Our results demonstrate that DHYZ significantly inhibits memory impairment of APP/PS1 mice.DHYZ may reduce neuronal damage in the cortex and hippocampus by inhibiting pp65 and down-regulating the release of cytokines IL-1 and TNF-α.
