微针介导HGF基因表达治疗湿疹的实验研究 被引量:1

Experimental study on treatment of eczema in mice with plasmid pHGF delivered by microneedles

摘要 目的通过微针经皮递送携带肝细胞生长因子(HGF)重组质粒pHGF,探究微针介导HGF基因表达治疗湿疹的作用。方法制备筛选pHGF透明质酸可溶性微针贴并进行表观机械性能检测、皮肤插入染色试验及小动物活体成像。实验小鼠随机分为模型组、阳性对照组、微针贴组及皮下注射组,采用2,4-二硝基氯苯(DNCB)皮肤致敏建立湿疹模型,按皮肤症状评分和苏木素-伊红(HE)染色切片观察皮肤病理情况,评价载药微针贴的治疗效果。结果透明质酸可溶性微针贴结构完整,机械强度较好,能较好地插入小鼠皮肤成功染色并能将质粒DNA有效递送到皮内表达。第28天后与模型组BALB/c小鼠相比,微针贴组和皮下注射组皮损积分较低(均P<0.01),HE病理染色切片显示,皮肤炎性浸润较轻,皮下附件修复较好。结论微针经皮递送pHGF基因表达能对湿疹模型小鼠起到较好的治疗作用。 Objective To investigate the effect of treating eczema with microneedle delivery of the recombinant plasmid pHGF carrying a hepatocyte growth factor(HGF).Methods The hyaluronic acid dissolving microneedle patches loaded with pHGF were prepared and screened for an apparent mechanical properties test,skin insertion staining test and in vivo imaging of small animals.The mice were randomly divided into four groups:the model group,positive control group,microneedle patch group and subcutaneous injection group.An eczema model was established by DNCB sensitization,and the therapeutic effect of the pHGF microneedle patches was assessed according to the score of skin lesions and hematoxylineosin(HE)staining section.Results The hyaluronic acid dissolving microneedle patches had a complete structure and good mechanical strength.The dissolving microneedle patches could be properly inserted into the mouse skin for staining and deliver plasmid DNA into the dermis for expression.After 28 days,compared with BALB/c mice in the model group,the microneedle patch group and subcutaneous injection group had lower skin lesion scores(P<0.01).HE pathological staining showed less inflammatory skin infiltration,and better repair of subcutaneous appendages.Conclusion The transdermal delivery of pHGF by microneedles could have a good therapeutic effect against eczema in mice.
