高效鉴别松毛虫赤眼蜂和玉米螟赤眼蜂寄生卵的微卫星引物筛选

Selection of SSR Primers for Efficient Identification of Parasitized Eggs between Trichogramma dendrolimi Matsumura and Trichogramma ostriniae Pang et Chen

摘要 为研究松毛虫赤眼蜂和玉米螟赤眼蜂对玉米螟卵的寄生情况,筛选了可用于早期鉴定和区分2种赤眼蜂的微卫星引物,并对被2种赤眼蜂寄生后12 h的玉米螟卵进行测试。结果表明:从设计的10对微卫星引物中,筛选出2对引物可用于鉴定松毛虫赤眼蜂和玉米螟赤眼蜂,其中1对可用于对同一寄生卵内赤眼蜂种类鉴定和区分;凡只能扩增出松毛虫赤眼蜂或玉米螟赤眼蜂特异性条带的卵,均被单独寄生,同时扩增出2种赤眼蜂混合条带的卵则被共寄生。此外,所有检测混合接蜂12 h后的玉米螟卵块中均存在2种蜂的共寄生卵,表明2种蜂之间存在共寄生现象。说明利用微卫星分子标记技术可以实现对赤眼蜂早期种类的鉴定,并为进一步开展松毛虫赤眼蜂和玉米螟赤眼蜂卵寄生关系的研究和优势蜂种筛选提供高效的检测方法和理论依据。 In order to illustrate the parasitic relationship between Trichogramma ostriniae and Trichogramma dendrolimi on eggs of Ostrinia furnacalis,ten pairs of microsatellite primers were selected to distinguish the two species.The eggs parasitized within 12 hours by two species were tested using microsatellite markers.The results showed that in total ten pairs of selected SSR primers,two pairs could be used to discriminate the species which parasitized the eggs alone either by T.dendrolimi or T.ostriniae,and one of them could be used to identify the eggs′multiparasitism by the two species.One specific band could be amplified for parasitized eggs which were considered to be parasitized by one species,and multiparasitism was confirmed through simultaneous amplification of mixed bands from one parasitized egg.Moreover,multiparasitism was observed between two species,while multiparasitized eggs could be detected in all parasitized eggs which were parasitized by two Trichogramma species within 12 hours.The results confirmed an efficient method for early detection of the Trichogramma species based on microsatellite markers,providing a theoretical basis for selected dominant parasitoids and multiparasitism characteristics for further study.
