摘要
目的观察亚砷酸钠对L-02肝细胞miR-21和Gadd45α表达的影响,并探讨其在砷致肝细胞损伤中的作用。方法以MTT法观察2.5~160μmol/L NaAsO2染毒6~48 h对L-02细胞活力的影响。以qRT-PCR法检测L-02细胞mi R-21的表达情况,采用免疫印迹法(Western blot)检测Gadd45α蛋白表达水平,并分析二者之间相关性。结果随着染砷剂量和时间的增加,L-02细胞活力逐渐降低,差异有统计学意义(P<0.05);miR-21相对表达量逐渐增加,且各剂量染毒组均高于对照组,差异有统计学意义(P<0.05);Gadd45α蛋白相对表达表现为先增高后降低的趋势,差异有统计学意义(P<0.05)。相关性分析结果显示,染毒不同剂量NaAsO2的L-02细胞miR-21 mRNA表达与Gadd45α蛋白表达存在正相关(r=0.545,P=0.036);染毒不同时间NaAsO2的L-02细胞miR-21 mRNA表达与Gadd45α蛋白表达存在正相关(r=0.872,P=0.000)。结论砷致肝细胞损伤的机制可能与miR-21和Gadd45α的表达增高有关。
Objective To investigate the effects of sodium arsenite on the expression of miR-21 mRNA and GAdd45α protein in L-02 hepatocytes and their role in arsenic-induced liver injury. Methods The cell viabilities of L-02 hepatocytes exposed to 2.5-160 μmol/L NaAsO2 for 6-48 h were determined by MTT assay. L-02 cells were exposed to 5-40 μmol/L NaAsO2 for24 h,and to 20 μmol/L NaAsO2 for 12-48 h respectively;The expression of mi R-21 in L-02 cells was detected by qRT-PCR,and the expression of Gadd45α was detected by Western blot,their correlation was analyzed also. Results With the increase of arsenic exposure dose and time,the activity of L-02 cells gradually decreased with statistically significant difference(P<0.05);The relative expression of miR-21 gradually increased,and showed significantly higher levels in exposure groups compared with that in control group( P<0.05);The relative expression of Gadd45α protein showed a trend of first increasing and then decreasing,the difference was statistically significant(P<0.05). There were positive correlations between the expression of mi R-21 and GADD45α protein in L-02 cells exposed to NaAsO2 with different doses(r=0.545,P=0.036) and for different time(r=0.872,P=0.000). Conclusion Arsenic-induced injury in hepatocytes may be related to the increased expression of miR-21 and Gadd45α.
