摘要
目的研究慢性铝染毒对大鼠精子mtDNA拷贝数及血清相关指标的影响,探讨铝降低精子活力的可能机制。方法取96只清洁级雄性Wistar大鼠,随机分为高剂量组[1/5LD50,256.72 mg/kg]、中剂量组[1/10LD50,128.36 mg/kg]、低剂量组[1/20LD50,64.18 mg/kg]及对照组蒸馏水[0 mg/kg],每组24只,经饮水摄入Al Cl3,连续饲养16周。测定其精子活力及mtDNA拷贝数,同时检测大鼠血清Zn、Cu、MDA含量及SOD活力。结果各剂量的铝染毒组大鼠的精子活力、血清Zn含量及血清SOD活性均明显低于对照组(P<0.01),精子mtDNA拷贝数、血清Cu含量及MDA含量均明显高于对照组(P<0.01)。铝染毒剂量与精子活力呈负相关(P<0.001),与精子mtDNA拷贝数呈正相关(P<0.001);精子活力与精子mtDNA拷贝数呈负相关(P<0.001);精子mtDNA拷贝数与血清Cu、MDA含量正相关(P<0.001),与血清SOD活力呈负相关(P<0.001)。结论铝降低大鼠精子活力可能与精子mtDNA拷贝数的改变有关。
Objective To understand the effects of chronic aluminum exposure on sperm mtDNA expression and related indexes in serum,and to explore the possible mechanism of reduced sperm motility induced by aluminum.Methods A total of 96 male Wistar rats were randomly divided into four groups,high dose group(1/5 LD50,256.72 mg/kg),moderate dose group(1/10 LD50,128.36 mg/kg),low dose group(1/20 LD50,64.18 mg/kg)and the control group(0 mg/kg),24 in each group.The rats were treated with Al Cl3 through drinking water for 16 consecutive weeks.At the end of experiment,epididymal sperms were collected and sperm motility and sperm mtDNA copy number were determined.The serum Zn,Cu,MDA contents and SOD activity were also measured.Results The sperm motility,serum Zn content and serum SOD activity of the aluminum-treated groups were lower than those of the control group(P<0.01),the sperm mtDNA copy number,serum Cu content and MDA content were higher compared with the control group(P<0.01).The dose of aluminum was negatively correlated with sperm motility(P<0.001),and positively correlated with sperm mtDNA copy number(P<0.001);Sperm motility was negatively correlated with sperm mtDNA copy number(P<0.001);Sperm mtDNA copy number was positively correlated with serum Cu and MDA levels(P<0.001),and negatively correlated with serum SOD activity(P<0.001).Conclusion Aluminum-induced sperm motility decease is related to changes in sperm mtDNA copy number.
