摘要
目的:观察抗CD40L单克隆抗体对OVA诱导的哮喘小鼠Th2炎症反应的影响。方法:18只野生型BALB/c小鼠随机分为PBS干预哮喘组、抗CD40L单克隆抗体干预哮喘组和正常对照组,每组6只;抗CD40L单克隆抗体干预哮喘组小鼠于第一次激发前尾静脉注射抗CD40L单克隆抗体500μg,并用等量PBS对照。分别采用HE和PAS染色观察肺组织炎症和气道杯状细胞增生;姬姆萨染色检测肺泡灌洗液(BALF)细胞总数和分类计数;ELISA法检测BALF中IL-4、IL-5和IL-13水平;小鼠肺功能仪测定小鼠气道高反应性。结果:抗CD40L单克隆抗体干预哮喘组病理检查肺组织炎症明显减少,抗CD40L单克隆抗体可以减少哮喘小鼠气管基底膜PAS+细胞(杯状细胞)数量(P<0.05),减少BALF中细胞总数、巨噬细胞和嗜酸粒细胞数量(P<0.05),降低BALF中IL-4、IL-5和IL-13水平(P<0.05),抑制哮喘小鼠气道高反应性(P<0.05或P<0.01)。结论:尾静脉注射抗CD40L mAb可以抑制OVA诱导的哮喘小鼠Th2炎症反应。
Objective:To investigate the effect of anti-CD40 L mAb on Th2 inflammatory responses in OVA-induced mouse models of asthma.Methods:Eighteen wild type(WT)BALB/c mice were randomly divided into control group(n=6),PBS treated asthma group(n=6)and anti-CD40 L mAb treated asthma group(n=6).Administration of anti-CD40 L mAb in anti-CD40 L mAb treated asthma group and PBS in PBS treated asthma group were carried out 1 h prior the first challenge.The airway inflammation was analyzed by lung histologic assay,while goblet cell hyperplasia,total cell count and differential cell count in bronchial lavage(BALF),the concentrations of IL-4,IL-5,and IL-13 in BALF,and the concentrations of OVA-specific IgE in the scrum were measured using HE,PAS staining,May-Grueuwald Giemsa,and ELISA,respectively.Airway hyperreactivity(AHR)was measured by lung function instrument.Results:anti-CD40 L treatment strongly suppressed inflammatory cell infiltration in the airways,decreased PAS-positive goblet cells in the respiratory tract epithelium(P<0.05),and reduced the frequency of total cells,eosinophils and macrophages in the BALF(P<0.05),as well as significantly lowered the concentrations of IL-4,IL-5,and IL-13 in the BALF(P<0.05)in the OVA-induced asthmatic mice,as compared with that respecively in PBS treatment group.Furthermore,the levels of OVA-specific IgE in the serum were markedly lower in OVA-induced asthmatic mice treated with anti-CD40 L treatment than those in PBS treatment group(P<0.05).In addition,more weakened RL to Mch was found in OVA-induced asthmatic mice with anti-CD40 L treatment,as compared with that respecively in PBS treatment group(P<0.05 or P<0.01).Conclusion:Administration of anti-CD40 L mAb can suppress Th2 inflammatory responses in OVA-induced asthmatic mice.
