阿尔茨海默病患者血清脑源性神经营养因子水平及其与认知功能的相关性研究 被引量:2

Level of serum brain-derived neurotrophic factor and its correlation with cognitive function in patients with Alzheimer’s disease

摘要 目的研究阿尔茨海默病患者血清脑源性神经营养因子(BDNF)水平及其与认知功能的相关性。方法选取青岛市、临沂市精神卫生中心收治的阿尔茨海默病(Alzheimer’s disease,AD)住院患者40例作为研究组,同时选取同期健康志愿者40例作为对照组。研究组和对照组均于入组后采用酶联免疫吸附法(ELISA)测定血清BDNF水平。研究组患者开始治疗前采用简易智力状态检查表(MMSE)进行认知功能的评定,应用药物(美金刚,起始5 mg/d,每周增加5 mg/d,渐增至最大剂量20 mg/d)治疗12周后,再次采用MMSE评定认知功能,用同样的方法测血清BDNF水平。比较研究组与对照组血清BDNF的水平;比较研究组患者治疗前后的血清BDNF水平及MMSE评分,探讨血清BDNF水平与MMSE评分的相关性。结果研究组患者治疗前血清BDNF水平低于对照组,差异有统计学意义(P<0.05)。经美金刚治疗后,研究组患者BDNF水平较治疗前明显提高,MMSE评分也高于治疗前(P均<0.05)。Peason相关性分析显示,阿尔茨海默病患者血清BDNF水平与MMS评分呈正相关(r=0.536,P<0.001)。结论阿尔兹海默病患者的BDNF水平较健康人群低,并且其水平与患者的认知功能呈正相关性。 Objective To study the level of serum brain-derived neurotrophic factor(BDNF)and its correlation with cognitive function in patients with Alzheimer’s disease.Methods Forty hospitalized patients with Alzheimer’s disease in Qingdao and Linyi mental health center were selected as the study group.At the same time,40 healthy volunteers were selected as the control group.The serum BDNF levels of the study group and the control group were measured by enzyme-linked immunosorbent assay(ELISA)at the time of enrollment.The cognitive function of the study group were assessed by simple mental state Checklist(MMSE)before treatment.After 12 week of treatment with drug(memantine,5 mg/d at the begiming,add 5 mg/d per week,and gradually increasing to the maximan dose of 20 mg/d),the cognitive function of the study group was assessed again with MMSE,and the serum BDNF level was measured with the same method.The levels of serum BDNF in the study group and the control group were compared.The levels of serum BDNF and the MMSE score in the study group were compared before and after treatment,and the correlation between the level of serum BDNF and the MMSE score was investigated.Results The level of serum BDNF in the study group was lower than that in the control group,and the difference was significant(P<0.05).After treatment with memantine,the serum BDNF level in the study group were significantly improved,and the MMSE score in the study group was significantly improved,compared with those before treatment(all P<0.05).Spearman correlation analysis showed that the level of serum BDNF was negatively correlated with the MMSE score in patients with Alzheimer’s disease(r=0.563,P<0.001).Conclusions The level of BDNF in patients with Alzheimer’s disease is lower than the healthy people,which is positively correlated with the cognitive function.
