摘要
目的:研究寻常型银屑病血热、血瘀、血燥证形成与Th分化、JAK/STAT信号通路关系。方法:收集寻常型银屑病患者33例(血热证15例、血瘀证10例、血燥证8例),正常组健康人10名,运用流式细胞术检测各组受试者外周血Th1、Th17细胞的分化比例差异,运用Western bolt法检测pSTAT4/STAT4、pSTAT3/STAT3表达差异,运用qPCR法检测T-bet、ROR-γt表达差异。结果:受试者外周血Th1、pSTAT4/STAT4、T-bet表达及Th17、pSTAT3/STAT3、ROR-γt表达互为正相关(P<0.01,P<0.05);血热证患者外周血Th1、Th17、pSTAT4/STAT4、pSTAT3/STAT3、T-bet、ROR-γt显著高于正常组(P<0.01),Th1血热证高于血燥证(P<0.05),Th17、pSTAT4/STAT4、pSTAT3/STAT3、T-bet、ROR-γt血热证高于血瘀证、血燥证(P<0.05,P<0.01)。结论:在Th细胞异常分化过程中,JAK/STAT信号转导通路的异常活跃可能是银屑病血热证形成的物质基础,且JAK/STAT信号通路可能与血热证向血瘀、血燥证转变的过程相关。
Objective:To observe the relationship between syndrome of blood heat,syndrome of blood stasis,syndrome of blood dryness psoriasis vulgaris in JAK/STAT signaling pathway and Th cell differentiation.Methods:A total of 33 psoriasis vulgaris patients(consist of 15 syndrome of blood heat,10 syndrome of blood dryness,8 syndrome of blood stasis)and 10 healthy participants were recruited.The ratio of Th1 and Th17 cells in peripheral blood of each group was detected by flow cytometry.The expression levels of transcription factor T-bet and ROR-γt in PBMCs were detected by real time-PCR method.The expression of pSTAT/STAT4 and pSTAT3/STAT3 expression in PBMCs were detected by Western bolt method.Results:The expression of Th1,pSTAT4/STAT4,T-bets and Th17,pSTAT3/STAT3,ROR-γt showed positive correlation with each other(P<0.01,P<0.05).Th1 and Th17 ratio,the expression levels of pSTAT4/STAT4,pSTAT3/STAT3,T-bet and ROR-γt were significantly higher in blood heat syndrome group than healthy participants(P<0.01).Th1 ratio in blood heat syndrome group was higher than blood drynesss syndrome group(P<0.05).Th17 ration and the expression levels of pSTAT4/STAT4,pSTAT3/STAT3,T-bet and ROR-γt in blood heat syndrome group were higher than blood dryness syndrome group and blood stasis syndrome group(P<0.05,P<0.01).Conclusion:In the pathological process of Th cell differentiation,the active status of JAK/STAT signaling pathway could be material basis in the formation of psoriasis blood heat syndrome.While the JAK/STAT signaling pathway may play an important part in blood heat syndrome truning into blood dryness syndrome and blood stasis syndrome.
