石斛多糖抑制自发性高血压大鼠晚期糖基化终产物受体mRNA的有效剂量观察 被引量:5

The effective dose of Caulis dendrobii polysaccharide on inhibiting receptor for advanced glycation end-product mRNA of spontaneously hypertensive rats

摘要 目的探讨石斛多糖抑制自发性高血压大鼠(SHR)晚期糖基化终产物(AGE)及晚期糖基化终产物受体(RAGE)水平的最低有效剂量,为临床药用石斛用量奠定基础。方法选取36只SHR随机分为对照组和治疗1~5组共6个组,每组6只,对照组给常温白开水,治疗1~5组分别给予石斛多糖药饮0.10 g/d(0.10 g组)、0.12 g/d(0.12 g组)、0.14 g/d(0.14 g组)、0.20 g/d(0.20 g组)、0.40 g/d(0.40 g组)治疗。疗程为2个月。于治疗前后观察RAGE mRNA表达变化情况,并与对照组比较。结果治疗前6组SHR的AGE和RAGE mRNA平均水平差异无统计学意义(P>0.05);治疗后0.12 g组、0.14 g组、0.20 g组和0.40 g组的AGE和RAGE mRNA平均水平低于对照组(P<0.01),也低于组内治疗前的水平(P<0.01)。治疗AGE和RAGE mRNA的最低有效剂量为石斛多糖0.12 g/d。结论石斛多糖可有效降低SHR体内AGE水平,降低RAGE基因的活化,对缓解SHR的血管重塑有积极的作用。治疗SHR的AGE和RAGE mRNA的最低有效剂量为石斛多糖0.12 g/d。 Objective To investigate the effective dose of caulis dendrobii polysaccharide on advanced glycosylation end products(AGE)and receptor for advanced glycation end-product(RAGE)in rats with spontaneously hypertensive(SHR)and its clinical significance.Methods Thirty-six SHR were randomly divided into Group Control and Group Treatment 1 to 5;in which boiled water,and caulis dendrobii polysaccharide medicine drink by 0.10 g/d,0.12 g/d,0.14 g/d,0.20 g/d and 0.40 g/d were given,respectively.Results There was no significant difference in SHR RAGE mRNA before treatment among the 6 groups(P>0.05).After treatment,the AGE RAGE mRNA levels in Group Treatment 2,3,4 and 5 were significantly lower than Group Control;and before those treatment(P<0.01).The minimum effective dose of caulis dendrobii polysaccharide on AGE RAGE mRNA was 0.12 g/d.Conclusion Caulis dendrobii polysaccharides can effectively reduce SHR and its advanced glycosylation end products,lower advanced glycosylation end-products of receptor gene activation,and reduce vascular remodeling.The minimum effective dose of caulis dendrobii polysaccharide is 0.12 g/d.
