摘要
目的探讨地参不同溶剂提取物中总多酚、总三萜含量及其抗氧化活性差异。方法采用水、无水乙醇、甲醇、60%乙醇、60%甲醇、60%乙腈和60%异丙醇等7种溶剂对地参进行提取,测定提取物中总多酚和总三萜的含量,进一步测定提取物还原力、羟基自由基清除率、1,1-二苯基-2-三硝基苯肼(1,1-diphenyl-2-picryl-hydrazyl,DPPH)自由基清除率和2,2’-联氮双(3-乙基苯并噻唑啉-6-磺酸)二铵盐[2,2’-azinobis-(3-ethylbenzthiazoline-6-sulphonate),ABTS]阳离子自由基清除率,评价其抗氧化活性。结果60%乙醇提取物中总多酚含量最高,为22.26 mg/g,无水乙醇提取物中总三萜含量最高,为0.74 mg/g。抗氧化活性结果表明,除无水乙醇提取物抗氧化能力较弱外,其余溶剂提取物均有较强的抗氧化能力,其中60%乙醇提取物的还原力、羟基自由基清除能力和ABTS阳离子自由基清除能力最强,60%乙腈提取物清除DPPH自由基效果最好,且其抗氧化能力与总多酚含量之间存在较高的正相关,与总三萜含量有一定的负相关。结论60%乙醇提取物中总多酚含量最高,抗氧化活性最好,是良好的天然抗氧化剂来源。
Objective To investigate the differences of the content of total polyphenols,total triterpenoids and their antioxidant activities in different solvent extracts of Lycopus lucidus var.hirtus Regel..Methods The Lycopus lucidus var.hirtus Regel.was extracted by 7 kinds of solvents including water,anhydrous ethanol,methanol,60%ethanol,60%methanol,60%acetonitrile and 60%isopropyl alcohol,the content of total polyphenols and total triterpenoids in the extract were determined.Reducing ability,hydroxyl radical,1,1-diphenyl-2-picryl-hydrazyl(DPPH)radical and 2,2’-azinobis-(3-ethylbenzthiazoline-6-sulphonate)(ABTS)cation radical scavenging rates of the extract were further determined to evaluate its antioxidant activity.Results The content of total polyphenols in 60%ethanol extract was the highest(22.26 mg/g),and the content of total triterpenoids in anhydrous ethanol extract was the highest(0.74 mg/g).The results of antioxidant activity showed that,except the anhydrous ethanol extract had weak antioxidant capacity,the other solvent extracts had strong antioxidant capacity,60%ethanol extract had the strongest reducing power,hydroxyl radical scavenging ability and ABTS cation radical scavenging ability,60%acetonitrile extract had the best DPPH radical scavenging ability.The antioxidant capacity was positively correlated with the content of total polyphenols,and negatively correlated with the content of total triterpenoids.Conclusion The 60%ethanol extract has the highest content of total polyphenols and the best antioxidant activity,which is a good source of natural oxidants.
