摘要
目的:研究黑碳颗粒(BC)对肺上皮细胞衰老的影响,以及西施舌提取物对衰老肺上皮细胞的保护作用与可能机制.方法:分离、纯化新生SD大鼠的肺上皮细胞,将细胞分为3组,分别为正常组、BC刺激组、BC刺激+西施舌提取物保护组.RT-qPCR法检测各组细胞P16与Rb mRNA的表达水平,Western blot法(蛋白质印迹法)检测各组细胞P16和Rb的蛋白表达水平.结果:BC刺激组肺上皮细胞P16与Rb mRNA和蛋白表达均较正常组增高(P<0.05);西施舌提取物保护组肺上皮细胞P16与Rb mRNA和蛋白表达均较BC刺激组下降(P<0.05).结论:BC刺激能加速肺上皮细胞的衰老,西施舌提取物能延缓这种衰老的程度,两者的作用机制可能与P16/Rb细胞信号转导通路有关.
Objective:To determine the effect of black carbon granules(BC)on lung epithelial cell aging and the protective effect of surf clam shell(Coelomactra antiquata)extract on aging lung epithelial cells,as well as the possible mechanism of action.Methods:Pulmonary epithelial cells were isolated and purified from neonatal SD rats.Three groups of culture cells were used:normal,BC stimulation,and BC stimulation+extract from Coelomactra antiquata.The levels of P16 and Rb mRNA expression were determined using RT qPCR,and the levels of P16 and Rb protein expression were determined using Western blot.Results:P16 and Rb mRNA and protein expression levels in lung epithelial cells were significantly higher in the BC stimulated group than those in the normal group(P<0.05);P16 and Rb mRNA and protein expression levels in lung epithelial cells in the Coelomactra antiquate extract protection group were significantly lower than those in the BC stimulation group(P<0.05).Conclusions:BC stimulation accelerates the aging of lung epithelial cells,whereas extract from Coelomactra antiquata retards the aging process.The mechanism may be related to the P16/RB cells signal transduction pathway.
