摘要
目的 观察硒酵母胶囊对自身免疫性甲状腺炎大鼠的抗氧化能力和免疫失衡的调节作用.方法 75只SD大鼠随机分为空白对照组、模型组、硒酵母胶囊低剂量组、硒酵母胶囊中剂量组、硒酵母胶囊高剂量组,每组15只.灌胃治疗1个月后,采用化学发光微粒子免疫法(CMIA)检测大鼠腹主动脉血中甲状腺过氧化物酶抗体(TPOAb)、甲状腺球蛋白抗体(TGAb)、促甲状腺激素(TSH)含量变化;采用酶联免疫吸附法(ELISA)测定动物血清中超氧化物歧化酶(SOD)、丙二醛(MDA)及谷胱甘肽过氧化物酶(GSH-Px)因子的含量;采用流式细胞术测定脾脏中CD4+CD25+Foxp3+含量;采用Western blotting法检测大鼠甲状腺组织中NF-κB蛋白的表达.结果 与模型组相比,硒酵母中高剂量组TSH、TGAb、TPOAb含量显著偏低(P<0.05);与模型组相比,硒酵母中高剂量组SOD与GSH-Px含量显著升高,MDA含量显著降低(P<0.05);与模型组相比,硒酵母高剂量组中CD4+CD25+Foxp3+/CD4+的比例值显著升高(P<0.05);与模型组相比,硒酵母中高剂量组中NF-κB蛋白含量显著偏低(P<0.05).结论 硒酵母胶囊可加强自身免疫性甲状腺炎大鼠抗氧化能力及免疫功能.
Objective To determine the effects of selenium yeast capsules on the antioxidative capacity and immune imbalance in rats with autoimmune thyroiditis.Methods Seventy-five SD rats were randomly divided into blank control group,model group,low-dose selenium yeast capsule group,medium-dose selenium yeast capsule group and high-dose selenium yeast capsule group(n=15 each).At 1 month after intragastric administration,the levels of thyroid peroxidase antibody(TPOAb),thyroglobulin antibody(TGAb)and thyroid stimulating hormone(TSH)in the abdominal aortic blood were measured by chemiluminescent microparticle immunoassay(CMIA).The levels of superoxide dismutase(SOD),malondialdehyde(MDA)and glutathione peroxidase(GSH-Px)factors in the serum were measured by enzyme-linked immunosorbent assay(ELISA).The level of CD4+CD25+Foxp3+in the spleen was measured by flow cytometry.The expression of NF-κB protein in the thyroid tissue was examined by Western blotting.Results Compared with the model group,the levels of TSH,TGAb and TPOAb in the medium-dose and high-dose selenium yeast capsule groups were significantly lower(P<0.05).Compared with the model group,the levels of SOD and GSH-Px in the medium-dose and high-dose selenium yeast capsule groups significantly increased,whereas the level of MDA significantly decreased(P<0.05).Compared with the model group,the ratio of CD4+CD25+Foxp3+/CD4+in the high-dose selenium yeast capsule group significantly increased(P<0.05).Compared with the model group,the protein level of NF-κB significantly decreased in the medium-dose and high-dose selenium yeast capsule groups(P<0.05).Conclusion Selenium yeast capsule may enhance the antioxidant capacity and immune function in rats with autoimmune thyroiditis.
