摘要
目的观察RP多糖在体外对HBV的抑制作用。方法 RP多糖设不同浓度组与HepG2.2.15细胞混合培养,同时设3-TC组和细胞对照组,采用CCK-8法检测药物对HepG2.2.15细胞的抑制作用,9d后通过ELISA法检测细胞培养液中的HBsAg、HBeAg含量,用FQ-PCR检测培养液中的HBV-DNA含量。结果药物浓度在小于1mg/mL时无明显细胞毒性;在各稀释浓度对HBsAg、HBeAg的分泌具有一定抑制作用,最大抑制率分别为50.9%和44.2%;HBsAg、HBeAg的治疗指数分别大于14.66%、12.39%。药物可抑制HBV-DNA的复制(<0.05)。结论 RP多糖在体外具有抗HBV作用。
Objective To observe the in-vitro inhibitory ef ect of RP Polysaccharide on HBV.Methods RP Polysaccharide groups with various concentrations were co-cultivated with HepG2.2.15 cells,while 3-TC groups and control groups were set for comparison.Then CCK-8 method was used to detect the inhibitory ef ect of drugs on HepG2.2.15 cells.Nine days later the concentrations of HBsAg、HBeAg in the cellculture fluid were tested with ELISA method,and the HBV-DNA content in the fluid was detected by Real-time FQ-PCR.Results RP Polysaccharide had no obvious celltoxicity at the concentration of thinner than 1mg/mL,and had inhibitory ef ect on the secretion of HBsAg、HBeAg at any level of diluted concentration,with maximum inhibition ratio 50.9%and 44.2%respectively.And the therapeutic index of HBsAg was more than 14.66%,while HBeAg,more than 12.39%.There was certain inhibition on proliferation of HBV-DNA(<0.05).Conclusion RP Polysaccharid can inhibit replication of HBV in Vitro.
