摘要
目的观察金雀异黄素对正常妇女成骨细胞增殖和分化的影响。方法采用四甲基偶氮唑蓝法,检测应用不同浓度金雀异黄素刺激成骨细胞增殖的情况,半定量RT-PCR及免疫组化法测定c-fos、c-jun基因及蛋白表达。结果以浓度为0mol/L的金雀异黄素为对照,10-10、10-8、10-6mol/L金雀异黄素增加人成骨细胞增殖达9.6%、14.4%、23.0%;金雀异黄素促进成骨细胞c-fos、c-jun基因及其蛋白的表达,表达呈剂量依赖性。结论金雀异黄素可刺激成骨细胞增殖,增加c-fos、c-jun基因及其蛋白的表达。
Objective To study the effects of genistein on Proliferation and differentiation of human normal osteoblasts. Methods Cell proliferation was tested by3-[4,5-Dimethy lthiazolzyl]-2.5-Diphenyl Tetrazolium Bromide(MTT) assay. The expression of c-fos and c-jun was measured by semi-quantitative reverse transcription-polymerase chain reaction (RT- PCR)and immuno-cytochemistry. Results Compared to the control group, genistein at concentrations of 10-10mol/L、10- 8mol/L and 10-6mol/L increased the proliferation of human osteoblasts by 9.6%、14.4% and 23.0% respectively, genistein upregulated c-fos and c-jun genes and protein expression in adose-dependent manner. Conclusions genistein stimulate proliferation of osteoblasts and increase the expression of c-los and c-jun.
