蛋白激酶C抑制剂灯盏花素对低氧预适应小鼠皮层NO/cGMP信号转导系统的影响

摘要 目的探讨NO/cGMP信号转导系统在低氧预适应中的作用及其与蛋白激酶C(Protein kinase C,PKC)的关系。方法将Blb/c近交系小鼠随机分为空白对照组(H0组)、低氧对照组(H1组)、低氧预适应组(H4组)、灯盏花素组(EB)和药物对照组(C)。观察:①H1,H4,EB和C组小鼠低氧耐受时间;②用分光光度计法测定各组小鼠皮层NOS活性、NO产量,放射免疫法测定cGMP含量。结果①H4,EB和C组小鼠低氧耐受时间显著多于H1组,且EB组明显多于H4组,(P<0.01);H4和C组没有显著差异。②H1,H4,EB和C组NOS活性、NO、cGMP含量均高于H0组,H4,EB和C组明显低于H1组,且EB组显著低于H4组,(P<0.01);H4和C组差异无显著性。结论NO/cGMP信号转导系统在低氧预适应中起重要作用,并且PKC能促进NO/cGMP信号转导系统的激活,PKC抑制剂灯盏花素可能通过抑制NO/cGMP信号转导系统参与低氧预适应的产生。 Objective To explore the role of NO/cGMP signaling pathway in hypoxic preconditioning and the its relation with protein kinase C(PKC).Methods Blb/c inbred strain mice were randomly divided into five groups:normal control group(H0),hypoxic control group(H1) and hypoxic preconditioning group(H4),erigeron breviscapus group(EB) and drug control group(C).Hypoxia tolerance time of H1,H4,EB and C group were observed.NOS activity and NO production of the cortex were measured by spectrophotometry and cGMP level measured by radioimmunoassay.Results ①Hypoxia tolerance time of H4,EB and C group increased significantly than that of H1 group,and the time of EB group even increased than that of H4 group(P<0.01).There was no significant difference between H4 and C group.②NOS activity,NO production and cGMP level of the cortex were significantly increased in H1,H4,EB and C group as compared with those of group H0.NOS activity,NO production and cGMP level of the cortex were significantly decreased in H4,EB and C group than those of group H1,and NOS activity,NO production and cGMP content of the cortex of EB group even decreased than those of H4 group(P<0.01).There was no significant difference between H4 and C group.Conclusion NO/cGMP signaling pathway may play an important role in the hypoxic preconditioning.PKC can promote the activation of NO/cGMP signaling pathway.PKC inhibitor erigeron breviscapus may be involved in the hypoxic preconditioning by inhibiting the activity of NO/cGMP signaling pathway.
