摘要
[目的]为了解龙岩市O157大肠杆菌人的感染、食品污染以及动物和媒介昆虫带菌情况。[方法]1999~2005年采集腹泻病人粪便、动物粪便以及各类食品、苍蝇、养殖场污水等标本,mEC增菌后用SMAC进行分离,纯化菌株经生化鉴定、血清分型、毒力及特征基因检测等。[结果]共检测2212份标本,检出O157大肠杆菌29株,阳性率为1.31%。其中猪粪为3.42%,牛粪为2.62%,牛肉为2.33%,鸡肉为2.06%,养殖场污水为2.27%。其余标本均未检出目的菌。O157∶H718株,O157∶NM6株,O157∶H?5株。对12株O157∶H7,进行了rfbO157和fliCH7基因的测定,结果rfbO157均为阳性,而fliCH7均为阴性。对18株(O157∶H714株、O157∶NM3株、O157∶H?1株),进行了SLT2、SLT1、eaeA、hly4种毒力基因检测,结果仅2株O157∶H7eaeA+hly阳性,1株O157∶H7hly阳性,其余菌株均为阴性。检出菌株对环丙沙星、氟嗪酸、丁胺卡那、庆大霉素敏感。检出ESBLs2株。[结论]我市畜、禽中存在O157大肠杆菌,未检出含有SLT2、SLT1毒素的菌株,未发现O157大肠杆菌病例和食物中毒患者,应加强宿主动物O157大肠杆菌监测。
[Objective]To understand the infection situation among human being,food contamination situation,and germ-carrying situation of animal and medium insect with Escherichia.coli O157 in Longyan City.[Methods]The different kinds of specimen were collected from 1999 to 2005,such as the feces of diarrhea patient,animals and different kinds of food,flies,dirty aqua of breed aquatic field.All specimens were enriched with mEC,then Escherichia.coli O157 was isolated with SMAC,the purified bacteria were detected by biochemical and serological methods including the virulence and characteristic gene detection.[Results]29 strains of Escherichia.coli O157 were separated from 2 212 specimen,the positive rate was 1.31%.The positive rates of the feces of pig,cattle were 3.42%,2.62% respectively,the positive rates of the beef,chicken were 2.33%,2.06% respectively,the positive rate of dirty aqua of breed aquatic field was 2.27%.Escherichia.coli O157 was not found in the rest specimen.18 strains were O157∶H7 type,6 strains were O157:NM type,5 strains were O157:H?type.Gene of O157∶H7,rfbO157 and fliCH7 were detected for 12 strains,the result was that the rfbO157 was positive and the fliCH7 was negative.SLT_2、SLT_1,eaeA and hly toxic genes were detected for 18 strains,the result was 2 strains of O157∶H7 carried eaeA and hly genes,1 strain of O157∶H7 carried hly gene,the rest were negative.All the strains were senstitive to Ciprofloxaxin,Ofloxacin,Amikacin and Gentamicin.2 strains of ESBLs was detected out.[Conclusion]Escherichia.coli O157 was found in domestic animals and birds in Longyan City,but the strains that carried SLT_2、and SLT_1 genes were not detected out.The cases and food poisoning caused by EHEC O157 were not found,the surveillance on Escherichia.coli O157 in host animal should be strengthened.
