高糖刺激肾小球系膜细胞Bcl-2、Smac、Fas的表达研究

摘要 目的:研究高糖刺激肾小球系膜细胞后,凋亡相关基因Bcl-2、Smac、Fas的表达情况。方法:将体外培养的大鼠肾小球系膜细胞分为正常对照组(糖浓度为5.6 mmol/L)和高糖组(糖浓度为30 mmol/L),采用四甲基偶氮唑蓝(MTT)比色法检测不同作用时间(12 h、24 h、36 h、48 h)下,系膜细胞的增殖状况,且在各时间点,采用免疫荧光染色法观察Bcl-2、Smac、Fas的表达情况并测定其荧光强度。结果:MTT比色法检测结果显示高糖组较对照组系膜细胞增殖明显,与作用时间呈正相关。同时,免疫荧光检测结果显示高糖组Bcl-2在以上不同时间作用下,与对照组相比,荧光强度增强(P<0.05),且高糖组随作用时间的延长,强度也相应增强,24 h、36 h、48 h各时间点的荧光强度与12 h相比,差异有统计学意义(P<0.05)。而Smac、Fas的荧光强度与对照组相比则减弱(P<0.05),且随作用时间的延长,强度也逐渐减弱,24 h、36 h、48 h各时间点与12 h相比,差异有统计学意义(P<0.05)。Bcl-2的荧光强度与Smac或Fas的荧光强度之间存在明显的负相关关系(P<0.05)。Smac的荧光强度与Fas的荧光强度之间存在明显的正相关关系(P<0.05)。结论:高糖刺激系膜细胞后,细胞增殖明显,Bcl-2的表达增加,Smac、Fas的表达下降,呈时间依赖性。 Objective:To investigate the expression of apoptosis gene associated with Bcl-2,Smac,Fas in glomeruli mesagi- nal (Mcs) cells under high concentration glucose.Methods:Mcs cells were divided into two groups:control group (with normal concentration glucose,5.6 mmol/L),high glucose group (with high concentration glucose,30 mmol/L).Using MTT assay,the proliferation of Mcs after 12 h、24 h、36 h、48 h was examined respectively.The expression of Bcl-2、Smac、Fas in Mcs cells was observed by fluorescence microscope at ...
