摘要
目的:探讨12-氧-十四烷酰佛波醇-13-乙酸酯(TPA)作用于食管癌细胞EC9706后,对NDRG1mRNA表达、细胞增殖、细胞周期的影响。方法:采用50nmol/L的TPA作用于EC9706细胞,MTT法检测细胞增殖情况,流式细胞仪检测细胞周期的变化,原位杂交法检测NDRG1mRNA水平。结果:TPA能上调NDRG1mRNA水平,抑制EC9706细胞增殖,细胞周期发生G1→S期阻滞。结论:TPA可能通过上调NDRG1mRNA的表达而抑制EC9706增殖,促进其分化。
Objective:To study the expression of NDRG1 mRNA,proliferation and cell cycle distribution of human esophageal carcinoma EC9706 cells after being treated with TPA.Methods:EC9706 cells were treated with 50nmol/L TPA,detecting the cell proliferation by MTT method and observing cell cycle distribution by Flow-cytometry,in situ hybridization technique to detect the NDRG1 mRNA level.Results:TPA could inhibit the proliferation of EC9706 cells,bring about G1 phase arrest and up regulate the expression level of NDRG...
