反义CⅡTA基因对鼠异位气管移植慢性免疫排斥反应的抑制作用

摘要 目的:构建携带鼠MHCⅡ类分子反式激活因子(MHC classⅡmolecule transactivator,CⅡTA)反义RNA的腺病毒载体,观察腺病毒介导该基因对小鼠异位气管移植慢性免疫排斥反应的抑制作用。方法:设计鼠CⅡTA mRNA为模板的反义片段,构建可表达反义片段的腺病毒载体(AdEasy-1系统),经HEK293细胞包装产生含反义片段重组腺病毒Ad-asCⅡTA,扩增、纯化。建立小鼠异位气管移植模型,用包装后的腺病毒注射并感染移植体周围,观察其对免疫排斥反应的抑制作用。结果:成功包装出含CⅡTA反义基因的重组腺病毒;重组腺病毒可感染气管移植体,Ad-asCⅡTA感染组移植气管组织炎症反应和纤毛上皮变性坏死等较对照组明显减轻。结论:重组腺病毒表达的小鼠反义CⅡTA基因能通过对MHCⅡ类分子的抑制而抑制异位气管移植的慢性免疫排斥反应。这为气管移植治疗气管肿瘤和气管狭窄等疾病奠定了一定的实验基础。 Objective:To construct adenovirus plasmid carrying MHC classⅡ molecule transactivator(CⅡTA)antisense RNA,and to observe the inhibitory effects of the gene on chronic immunologic rejection of heterotropic transplantation of trachea in mice.Methods: The antisense fragment with CⅡTAmRNA as nuclear plate was designed.The adenovirus plasmid carrying the antisense fragment(AdEasy-1 system) was constructed.After packaging with HEK293 cells,the recombinant adenovirus(Ad-asCⅡTA) was prepared.Then the mouse model of ...
