普罗布考对高脂血症大鼠肝脂质过氧化的影响及机制 被引量:5

摘要 目的研究普罗布考对高脂高胆固醇饮食诱导的大鼠肝脂质过氧化的影响及机制。方法将30只SD雄性大鼠随机分为对照组、高脂组和高脂普罗布考组,每组各10只。采用酶比色法等检测试剂盒检测大鼠血浆脂质水平,用硫代巴比妥酸法测定肝组织丙二醛(MDA)的含量,用比色法测定肝组织匀浆中的超氧化物歧化酶(SOD)及总抗氧化能力(T-AOC)的水平,用RT-PCR检测大鼠肝脏PON1 mRNA的表达。结果与对照组比较,高脂组大鼠的血脂水平增高,肝组织匀浆MDA的水平显著增高,SOD及T-AOC的水平则降低(P<0.05);而与高脂组比较,普罗布考组血脂水平及肝组织匀浆中MDA的水平则有明显的下降,肝组织中SOD及T-AOC水平则增高(P<0.05);PCR检测结果显示高脂组大鼠肝脏PON1 mRNA的表达下调(P<0.05),而普罗布考处理组的PON1 mRNA的表达上调(P<0.05)。结论普罗布考可以抑制高脂高胆固醇饮食诱导的大鼠肝脏脂质过氧化,上调肝脏PON1的表达。普罗布考对PON1的影响可能是其抗氧化作用的机制之一。 Objective To investigate the effects and mechanism of Probucol on lipid peroxidation of liver in hyperlipidemic rats.Methods30 rats were divided into 3 groups(control group, high fat group and high fat-probucol group, n=10, each group) in random.Serum triglycertide(TG), total cholesterol(TC), low density lipoprotein cholesterol(LDL-C) were determined by commercially provided kits.The level of total antioxidative capacity(T-AOC), superoxide dismutase(SOD) activity and malondialdehyde(MDA) were valued by comm...
