大鼠内毒素休克模型中脑红蛋白的表达变化及临床意义 被引量:3

摘要 目的探讨内毒素休克大鼠模型的额叶皮质、脑脊液(CSF)和血浆中脑红蛋白(Ngb)的表达变化及其意义。方法70只SD大鼠随机分为对照组和内毒素干预组。内毒素干预组经大鼠尾静脉注射内毒素16mg/kg,对照组注入等容量0.9%氯化钠溶液。于注射内毒素后3、6、12、24、48和72h收集血浆、CSF,并处死大鼠留取额叶皮质。应用酶联免疫吸附法、免疫印迹法和免疫组织化学法检测Ngb的表达变化;应用干燥法检测额叶皮质含水量。结果(1)酶联免疫吸附法显示,注射内毒素后额叶皮质、CSF、血浆中Ngb表达水平与对照组比较,差异有统计学意义(P<0.01);(2)免疫印迹法显示各组均表达相对分子质量约为17kD的Ngb蛋白。内毒素干预各组Ngb蛋白表达水平与对照组比较,差异有统计学意义。(3)Ngb免疫组化反应阳性物质主要位于神经元的细胞质,呈黄褐色。(4)注射内毒素后,脑组织含水量与对照组间差异均有统计学意义(P<0.01),48h和72h的含水量达峰值。结论在内毒素所致脑损伤中Ngb表达上调,且与内毒素的注入时间相关,Ngb表达上调是机体内源性神经保护机制之一。 Objective To study the changes of Cortex of frontal lobe,cerebrospinal fluid(CSF) and neuroglobin (Ngb) in rat endotoxic shock models.Methods Seventy adult SD rats were divided into operation groups (n=60) and control(n=10).Operation groups were given via vena caudalis an injection of endotoxin in a dose of 16 mg/kg,and control given equal volume of 0 9% sodium chloride solution.Plasma and CSF were collected at hours 3,6,12,24,48,72 after injection,and then rats were sacrificed and Cortex of frontal lobe was kept..The changes of Ngb expression were detected by enzyme linked immunosorbent assay(ELISA),Western blotting and immunohistochemical method,and water content of Cortex of frontal lobe by desiccation.Results ①ELISA showed that Ngb concentrations in plasma,CSF and cortex of frontal lobe were significantly higher in operation groups than in control after endotoxin injection(P<0.01),reaching peak at hour 48.②Western Blotting showed that 17 kD relative molecular mass of Ngb was observed in all groups.The Ngb expression,which was significantly higher in operation groups than in control,was the highest in groups of 48 h,72 h.③IHC showed that Ngb immunohistochemistry-positive products were mainly located in the cytoplasms of the neurons,being yellow-browny in color.④After endotoxin injection,the water contents of brain tissues were significantly higher than control (P<0.01),reaching peak at h 48(83.13%).Conclusion The up-regulation of Ngb is induced by endotoxic shock in rats,and changed in a time-dependent manner.It is one of the mechanisms of endogenous neuroprotection during endotoxic shock.
