摘要
目的:制备氟甲砜霉素(FF)的最佳人工抗原并鉴定。方法:采用琥珀酸酐将FF的羟基活化后制备出半抗原氟甲砜霉素半琥珀酸(FF-HS),并经质谱(MS)鉴定合成成功。采用碳二亚胺法将FF-HS与牛血清白蛋白(BSA)偶联,采用混合酸酐法将FF-HS与卵清白蛋白(OVA)偶联,采用羰基二咪唑法将FF直接与BSA偶联,制备了3种人工抗原(FF-HS-BSA、FF-HS-OVA和FF-BSA)。采用紫外吸收光谱分析和动物免疫实验(3种偶联物选择前2种分别作为免疫原与包被原进行抗血清制备及ELISA实验)鉴定3种人工抗原。结果与结论:紫外吸收光谱分析初步证实3种人工抗原合成成功。在动物免疫实验中FF-HS-BSA与FF-HS-OVA得到了效价较高的特异性抗体,IC50分别为150、300μg/L。FF-BSA免疫原免疫效果不理想。
