摘要
目的采用HPLC法测定小鼠脑匀浆中的盐酸尼莫司汀。方法使用Diamonsil C18柱,流动相为甲醇-水-三乙胺(20∶79.5∶0.5,磷酸调pH3),流速1.0mL·min-1,检测波长241nm,柱温35℃。结果盐酸尼莫司汀0.20~5.00mg·L-1与峰面积的线性关系良好(r=0.9999);平均方法回收率95.7%,平均萃取回收率101.2%,日内、日间RSD均<5%。结论该方法简便、灵敏、准确,可用于测定脑匀浆中的盐酸尼莫司汀。
OBJECTIVE To establish a method for the determination of nimustine hydrochloride in mouse brain homogenate.METHODS The concentration of nimustine hydrochloride was determined using HPLC-UV with the Diamonsil C18 column.The mobile phase was methanol-water-triethylamine (20∶79.5∶0.5,adjusted to pH3 with H3PO4).The flow rate was 1.0 mL·min -1.The detection wavelength was at 241nm,and the column temperature was 35 ℃.RESULTS The linear range was 0.20-5.00 mg· L -1(r = 0.9999).The average methodological recovery ...
