哈茨木霉菌Tri101基因的克隆与序列分析 被引量:2

摘要 根据拟枝孢镰刀菌等真菌中Tri101的保守序列设计简并引物,利用简并PCR和基因组步行法克隆了哈茨木霉菌Tri101基因.结果表明:Tri101基因的开放阅读框为1 314 bp,无内含子存在,与预计的Tri101序列特征一致.该基因编码蛋白质由437个氨基酸组成,预测相对分子质量约为4.75 kD,系统进化树显示其与棒曲霉(Aspergillus clavatus)NRRL 1亲缘关系最近,与NRRL 1中3-O-乙酰转移酶的的蛋白质同源性达到55%.Blast数据库分析表明其属于转移酶总科,单端孢酶烯乙酰转移化酶.以禾谷镰刀菌(Fusarium graminearum)TRI101蛋白(Parent PDB:3b2s Chain:A)的晶体为模板进行了同源建模,为进一步研究该基因的功能奠定基础.
