高压氧对卡那霉素致聋肾虚豚鼠耳蜗琥珀酸脱氢酶的影响

The Effect of Hyperbaric Oxygen on Succinate Dehydrogenase of Kanamycin Induced Ototoxic Cochlear in Kidney-deficiency Guinea Pig

摘要 [目的]观察高压氧(HBO)对卡那霉素致聋肾虚豚鼠的耳蜗琥珀酸脱氢霉(SDH)变化的影响。[方法]将30只雄性豚鼠随机分为3组,每组10只,分别为正常对照组(正常组)、肾虚耳聋模型对照组(模型组)、肾虚耳聋高压氧治疗组(治疗组),以听性脑干反应(ABR)及SDH组织化学检测为指标。[结果]模型组豚鼠ABR反应阈明显升高,治疗组ABR反应阈升高幅度明显低于模型组,两组比较差异有统计学意义(P<0.05);模型组SDH显色变淡或消失,毛细胞受损显著,治疗组SDH变化、毛细胞受损程度均明显低于模型组。[结论]HBO能提高内耳SDH的活性,促进受损细胞功能恢复,对卡那霉素致聋肾虚型豚鼠有一定的治疗作用。 [Objective] To observe the effect of hyperbaric oxygen(HBO)on succinate dehydrogenase (SDH)of kanamycin induced ototoxic cochlear hair cells in kidney-deficiency guinea pig.[Methods]30 healthy male guinea pigs were divided randomly into three groups:normal group,model group and therapy group,10 in each group.Auditory brain stem response (ABR)and SDH in the cochlear hair cells were taken as indexes.[Results]The thresholds of ABR of the guinea pigs in the model group rose markedly,while those of therapy group rose slightly.The difference was significant between two groups.The degree of hair cell lesion in the model group was higher than those in the therapy group,and the change of SDH activity within cochlear hair cells in the model group was more obvious than those in the therapy group.[Conclusions]HBO therapy has curative effect on kanamycin induced ototoxic cochlear in kidney-deficiency guinea pig.The mechanism may be associated with its protected activity of SDH in inner ear and the recovery of impaired cell.
