摘要
目的比较三种分子生物学方法检测临床手足口病标本的敏感性、特异性及优缺。方法采用一步法逆转录-聚合酶链反应(RT-PCR)、巢式PCR和实时RT-PCR(Real-time RT-PCR,RRT-PCR)三种分子生物学方法检测临床手足口病标本,分别计算检测肠道病毒71型(EV71)阳性率差异的显著性。结果住院及重死病例以EV71感染为主,轻症病例以柯萨奇A组16型(CA16)感染为主;一步法RT-PCR检测EV71病原的阳性率与另外两种差异均有显著性,另外两种差异无显著性。结论巢式PCR及实时RT-PCR检测能显著提高检测的敏感性,一步法RT-PCR可以用在病毒株的鉴定及分析中。
