三种分子生物学方法诊断手足口病的比较 被引量:8

摘要 目的比较三种分子生物学方法检测临床手足口病标本的敏感性、特异性及优缺。方法采用一步法逆转录-聚合酶链反应(RT-PCR)、巢式PCR和实时RT-PCR(Real-time RT-PCR,RRT-PCR)三种分子生物学方法检测临床手足口病标本,分别计算检测肠道病毒71型(EV71)阳性率差异的显著性。结果住院及重死病例以EV71感染为主,轻症病例以柯萨奇A组16型(CA16)感染为主;一步法RT-PCR检测EV71病原的阳性率与另外两种差异均有显著性,另外两种差异无显著性。结论巢式PCR及实时RT-PCR检测能显著提高检测的敏感性,一步法RT-PCR可以用在病毒株的鉴定及分析中。 Objective In the light of detecting Hand-foot-and-mouth disease(HFMD) etiology in the future,compared the three types of molecular biology technologies on the sensitivity,specificity,advantage and disadvantage upon detecting the HFMD clinical specimens.Methods The clinical HFMD specimens were detected by three molecular biology technologies,which concluding One-step RT-PCR,Nest PCR and Real-time RT-PCR.In the same time compared their different in detecting EV71 positive rate.Results The inpatients or dying ...
