摘要
[目的]观察睡眠剥夺对大鼠饮食行为、能量消耗、体温和抗氧化能力的影响,探讨中药的干预作用及其抗氧化应激的机制。[方法]将雌性SD大鼠32只随机分为4组,即正常对照组、模型对照组、抗衰老片组、六味地黄丸组,每组8只,利用"小平台水环境法"建立大鼠睡眠剥夺模型,分别进行体温、体重、进食量和饮水量的监测,并计算日能耗,同时检测机体血清SOD活性和MDA含量。[结果]72h睡眠剥夺后,模型对照组大鼠体重、进食量和饮水量均明显低于正常对照组(P<0.05,P<0.01),日消耗能量和体温明显高于正常对照组(P<0.05,P<0.01),且机体血清SOD活性明显降低(P<0.01),MDA含量明显升高(P<0.01);进行抗衰老片和六味地黄丸干预后,均能有效预防睡眠剥夺后大鼠的体重、进食量、饮水量的降低,并能降低日消耗能量和体温,显著提高机体SOD活性(P<0.05,P<0.01),降低MDA含量(P<0.05,P<0.01)。[结论]睡眠剥夺可引起大鼠饮食行为的改变、耗能增加、体温升高,抗氧化应激能力下降。而抗衰老片和六味地黄丸等中药具有抗睡眠剥夺引起的氧化应激的作用,其机制可能与提高机体的抗氧化能力有关。
[Objective]To explore the effect of sleep deprivation on oxidative stress in rats,moreover,the effect of Anti-aging tablets was observed in rats of sleep deprivation.[Methods]A total of 32 female SD rats were randomly divided into four groups: normal control group,model control group,Anti-aging tablets and Liuwei Dihuang Pills group,8 rats per group.The 'flower pot' technique was used to establish rats sleep deprivation model.The body temperature,weight,intake food,drinking amount of water,daily consumption of energy,serum of SOD activity and MDA content of all rats were measured respectively.[Results]72h after sleep deprivation,compared with normal control group,the weight,intake food,and drinking amount of water were obviously decreased in model control group(P<0.05,P<0.01),daily consumption of energy,and body temperature were obviously increased(P<0.05,P<0.01),activity of serum SOD was obviously decreased(P<0.01),content of serum MDA was obviously increased in model control group(P<0.01).After administration of Anti-aging tablets and Liuwei Dihuang Pills,the extent of decreased weight,intake food,and drinking amount was mitigated,the daily consumption of energy,and body temperature were decreased,the activity of serum SOD was advanced,the content of serum MDA was reduced of all rats(P<0.05,P<0.01).[Conclusion]The study suggests that sleep deprivation can change the intake food and drinking amount of water,increase consumption of energy and rise body temperature,Anti-aging tablets and Liuwei Dihuang Pills can confront with oxidative stress in rats model with sleep deprivation on sub-health condition.And the mechanism may be related to the enhance ability of anti-oxidation.
