摘要
目的通过构建急性心肌梗死(Acute myocardial infarction,AMI)再灌注兔子模型,观察重组人脑利钠肽(Lyophiluzed Re-combinant Human Brain Natriuretic Peptide,rhBNP)对兔子心肌梗死再灌注损伤的保护作用。方法制备实验性心肌梗死再灌注损伤模型,随机分为 3 组 :A 组 :假手术组、B 组 :模型组、C 组 :rhBNP 组(治疗前组和治疗后组)。测定心肌酶学(血清天冬氨酸氨基转移酶AST、乳酸脱氢酶 LDH、磷酸肌酸激酶 CK)、超氧化物酶(supemxide dismutase,SOD)活性、髓过氧化物酶(myeloperoxidase,MPO)活性、丙二醛(malonaldehyde,MDA)含量以及治疗前后 N 端脑钠素前体(N-terminal pro-brain natriuretic peptide,NT-proBNP)、超敏 C反应蛋白(high-sensitivity CRP,hs-CRP ,hs-CRP)水平变化。结果与 A 组相比,B 组兔子的 SOD 活性明显下降,MDA 及 MPO 含量均明显增加(P<0.05 或 P<0.01);AST、LDH、CK-MB 均显著升高(P<0.01);与 B 组比较,C 组兔子 SOD 活性明显升高,MDA 及 MPO 含量均明显降低(P<0.05);AST、LDH、CK 活性明显降低(P<0.05 或 P<0.01);C 组治疗前与治疗后相比 NT-proBNP、hs-CRP 水平均显著改善(P<0.05),治疗后血清 NT-proBNP、hs-CRP 水平较治疗前下降较为明显(P<0.05)。结论 AMI 再灌注后静脉应用 rhBNP 对缺血梗死再灌注损伤有明显保护作用,对于急性心肌梗死患者来说,重组人脑利钠肽可作为又一种有效的治疗药物。
Object ive The study aimed to investigate the protect ive effect of lyophiluzed recombinant human brain natriuretic peptide against myocardial infarction reperfus ion injury(IRI) in rabbit .Meth ods: I/R model for rabbit was established,thirty rabbits were rondomly into the three gro ups(n=10,respect ively):control group、model group、rhBNP group(Before treatment group and after t reatment group). myocardial enzymes(level of aspartate trans amin ase、lactate degyd rogenase、creat ine ph osp hokinase)、the su pemx ide dismutas e(SOD)、myeloperox idas (MPO)、 malonaldehyde(MDA) 、N-terminal pro-brain natriuretic pept ide and high-sensit ivity were detected in different groups.Resu lts:the level ofAST、 LDH、CK-MB in gurop B were singni��cantly higher than the group A(P < 0.01), the level of AST、LDH、CK-MB in gurop C were singni��cantly lower than the group B(P < 0.05 or P < 0.01),SOD activity in group B was signi��cantly lower than the group A、the Content of MDA and MPO in group B increases obviously(P < 0.05 or P < 0.01),SOD activi ty in group C was s igni��cantly higher than the group B、the Content ofMDA and MPO in group C lower obviously(P < 0.05); compared with Before treatment group in gurop C,after t reatment ,the level of N-terminal pro-brain natriuretic peptide and high-sensi tivity were highly lower(P < 0.05). Con clusion :There was protective effect of intravenously rhBNP against myocardial IRI in rabbit in vivo, For patients with acutemyocardial infarction with speaking, rhBNP can be used as an effective drug treatment.
