摘要
以卵黄免疫球蛋白(immunoglobulin yolk,IgY)为配基,对小牛肠组织来源的碱性磷酸酶进行免疫亲和纯化。采用1.5mol·L-1的NaCl进行洗脱,可使酶的纯化倍数达到64倍,活力回收率达到90%。IgY抗体对碱性磷酸酶亲和力的测定结果显示固有亲和力和相对亲和力分别为6.275×10-7 mol·L-1和5.0 μg·ml-1。
Immunoglobulin yolk (IgY) was employed as immunoaffinity ligand to purify alkaline phosphatase from crude enzyme. After elution with 1.5mol·L-1 sodium chloride a 64-fold purified enzymatic preparation was obtained in one step with a yield of 90%. The absolute affinity and relative affinity of anti-AKP IgY to AKP are 6.275×10-7 mol·L-1 and 5.0 μg·ml-1,respectively.
