摘要
目的应用Aβ25-35孵育PC12细胞制作细胞损伤模型,探讨虫草素对PC12细胞损伤的保护作用.方法将不同组别PC12细胞用Aβ25-35诱导48 h后,采用MTT法测定细胞活力,LDH法测定细胞膜通透性,化学比色法测定细胞中SOD活性和MDA含量,Western blot法检测PC12细胞中Caspase-3的表达.结果与模型组相比,两种剂量的虫草素均使细胞存活率显著增加,细胞培养上清中LDH含量显著减少,细胞中SOD活性显著升高,MDA含量显著下降,并明显降低PC12细胞Caspase-3的表达.结论虫草素对Aβ25-35诱导的PC12细胞死亡、氧化损伤和细胞凋亡有明显保护作用.
Objective To investigate the protective effect of cordycepin on PC12 cells injury induced by Aβ25-35.Method Different groups of PC12 cells were treated with Aβ25-35 for 48 h,MTT assay was used to detect the viability of PC12 cells,the membrane permeability was measured by means of LDH method,the level of SOD and MDA were tested by using chromatometry,and western blot was used to evaluate Caspase-3 protein level.Results Compared with those of the model group,the cell viability was significantly increased after treatment of two doses of cordycepin.Cordycepin also decreased the LDH release from PC12,the activity of SOD was enhanced and content of MDA in PC12 was reduced by cordycepin.The protein level of Caspase-3 in cells was elevated after the cordycepin treatment.Conclusion Cordycepin has protective effects on oxidative damage and apoptosis of PC12 cells induced by Aβ25-35.
