摘要
目的通过月桂酸钠心肌内注射,建立微循环栓塞的动物模型,探讨月桂酸钠致微循环栓塞的病变机制以及对心脏功能的影响。方法将SD大鼠随机分为三组:模型组、假手术组和对照组。模型组大鼠经开胸,分离心脏和主动脉,在夹闭经主动脉的同时,向心尖部注入月桂酸钠,假手术组注入相等体积生理盐水,对照组不做任何处理。于术后7天、28天时取心肌制成切片,经HE与马休黄-酸性品红-苯胺蓝(改良Lendrum法)染色,观察微循环血栓的形成;透射电镜下观察血管内皮细胞超微结构的变化,术后28天应用超声心动图检测心脏功能的改变。结果模型组大鼠心肌微循环血管中,可见存在大量由血小板纤维蛋白和胶原构成的血栓,电镜下内皮细胞间紧密连接增宽,微血管挛缩狭窄,管腔通透性增大;超声检查示模型组左心室收缩末径显著大于假手术组(3.97±0.12:3.28±0.42,P<0.05),心功能明显降低(66.95±1.79:79.39±1.98,P<0.05);而假手术组和正常对照组未见明显变化。结论月桂酸钠心肌内注射可以有效的建立冠脉微循环栓塞的动物模型,导致心脏微结构及功能的改变,内皮化学性损伤是月桂酸钠导致微栓塞的机制。
Objective To explore the mechanism of microthrombus induced by sodium laurate in cardiac microcirculation in rats.Methods Total 80 SD rats were randomly divided into 3 groups:model group,sham group and control group.In model group,exposed and occluded the aorta while injected with sodium laurate into apical heart muscle.In sham group,took normal saline instead and no operation with control group.At the day 7 and 28 after the operation,in-situ microthrombosis was detected with HE and improved Lendrum staining.The ultrastructure changes in vascular endothelial cells were observed with transmission electron microscopy.Echocardiography was used to assess the cardiac function at day 28.Results The Microthrombus consisting of platelet ? bers and collagen could be observed mostly in model group;tight junctions between cells became wide and microvascular became narrowed under the transmission electron microscopy sight.The LVIDs in model group was signi? cantly greater than in the sham group(3.97±0.12:3.28±0.42,P<0.05) and the cardiac functions decreased dramaticly in the model group compared with control group(66.95±1.79:79.39±1.98,P<0.05).No signi? cant differences observed between sham group and control group.Conclusion Microthrombus can be successful induced by sodium laurate injection which can cause cardiac dysfunction.The injury of endothelial cells is attributed to this pathogenesis change.
