摘要
目的采用半定量反转录-聚合酶链反应(reverse transcription-polymerase chain reaction,RT-PCR)研究表皮生长因子(epidermal growth factor,EGF)及受体(epidermal growth factor receptor,EGFR)在大鼠胃溃疡自愈期间胃黏膜中表达的变化及其意义,并探讨半定量RT-PCR法用于此项检测的意义。方法大鼠胃前壁下注射冰乙酸制备胃溃疡模型,Redzol法分别提取正常组、胃溃疡术后1、4、6、10、14d组大鼠胃黏膜(每组6只)组织中总RNA,RT-PCR法对EGF mRNA、EGFR mRNA进行半定量分析。结果 RT-PCR法显示在胃黏膜愈合过程中EGF mRNA、EGFR mRNA表达逐渐增强,EGF mRNA在溃疡术后d 10表达最强,(1.99±0.10,P<0.01),之后稍减弱,EGFR mRNA在溃疡术后6d表达最强(1.69±0.10,P<0.01),之后减低。结论在胃溃疡自愈过程中,胃黏膜能内源性表达EGF mRNA、EGFR mRNA,促进黏膜的修复与愈合,同时半定量RT-PCR法用于此项检测具有快速、简便、特异性强的特点。
Objective To investigate the expression and significance of EGF mRNA、EGFR mRNA in gastric mucosa during gastric ulcer healing period by RT-PCR and discuss applications of RT-PCR.Methods The test model was built by injecting acetic acid in to submucosa of gastric anterior wall.The total RNAs of gastric mucosa were extracted from rats in normal group and gastric ulcer group(n=6)on 1 d,4 d,6 d,10 d,14 d by Redzol,while semi-quantitative analysis for the results of EGF mRNA、EGFR mRNA was made by RT-PCR.Results During the healing of gastric mucosa,expressions of EGF mRNA、EGFR mRNA increased gradully,but on the 10th d(1.99±0.10)and 6th d(1.69±0.10)after gastric ulcer,their expressions were the strongest respectively(P<0.01),and then attenuated gradually.Conclusion During the healing of gastric ulcer,gastric mucosa could endogenously express EGF mRNA、EGFR mRNA,and promote mucosa recovery,while semi-quantitative RT-PCR is a simple and sensitive method to detect RNA.
