金樱子诱导HO-1表达对高糖条件下LECs活性氧产生及线粒体膜电位的影响 被引量:3

Effects of Rosa Laevigata Michx Induced HO-1 Expression on ROS and Mitochondrial Membrane Potential of LECs Cultured in High Glucose Medium

摘要 目的:观察金樱子(Rosa laevigata Michx.,RLM)对高糖条件下人晶状体上皮细胞(lens epithelial cells,LECs)活性氧(reactive oxygen species,ROS)产生和线粒体膜电位(itochondrial membrane potential,MMP)变化的影响,同时探讨血红素氧合酶-1(Heme oxygenase-1,HO-1)在此过程中的作用。方法:将体外培养的人LECs系SRA01/04细胞分为6组:对照组、高糖组、RLM处理组(高糖+2.5、5.0、10.0 g/L RLM)、HO-1沉默组(高糖+HO-1沉默+RLM 10.0 g/L),用荧光探针法检测细胞中ROS,罗丹明123染色检测MMP,Western blot检测HO-1表达。结果:⑴高糖组LECs细胞ROS产生较对照组增加,RLM处理后ROS的产生量随着RLM浓度的增高而减少(P均<0.01),RLM浓度为10.0 g/L时,ROS含量接近对照组(P>0.05);⑵高糖组MMP水平低于对照组水平(P<0.01),RLM处理后MMP水平较高糖组升高,至10.0 g/L时MMP与对照组差异无统计学意义(P>0.05);⑶与高糖组比较,RLM组HO-1表达增加,而干扰HO-1表达后,相对于10.0 g/L RLM组,干扰组ROS产生增加,MMP水平降低(P均<0.05)。结论:RLM能在一定程度上抑制高糖诱导的人LECs中ROS产生及MMP改变,该过程可能与其诱导HO-1的表达有关。 Objective To observe the effects of RLM on the production of ROS and MMP levels in LECs cultured in high glucose medium,and to explore the possible role of HO-1. Methods Human LECs line SRA01 /04 cells were cultured and randomly divided into six groups,including control group,high glucose group,RLM groups( high glucose + 2. 5,5. 0,10. 0 g / L RLM),HO-1 silenced group( high glucose + HO-1 siRNA + 10. 0 g / L RLM). Then the fluorescence probe method,rhodamine staining and Western blot were adopted to detect the production of ROS,MMP levels and HO-1 expression,respectively. Results ⑴The ROS level in high glucose group was significantly higher than that of control group,but the ROS levels were obviously decreased in RLM groups following the increased concentrations( all P < 0. 01),when the concentration of RLM reached to 10. 0 g / L,the ROS level was almost recovered to the level of control group( P > 0. 05). ⑵The MMP level in high glucose group was significantly lower than that of control group,and it was increased after treated with RLM( all P <0. 01),when the concentration of RLM reached to 10. 0 g / L,the MMP level had no significant difference with that of control group( P > 0. 05). ⑶Compared with high glucose group,the expression of HO-1 was increased in RLM group. After the interference of HO-1 expression,the ROS production and MMP level of HO-1 silenced group were increased and decreased in10. 0 g / L RLM group,respectively( both P < 0. 05). Conclusion RLM can inhibit high glucose induced ROS production and MMP change in LECs,which is probably mediated by HO-1.
