摘要
目的观察肺动脉内皮细胞在低氧情况下TSP-1、TGF-β1及EGFR的表达。方法原代培养SD乳鼠肺动脉内皮细胞,传代后的细胞分为常氧对照组和低氧(3%氧浓度)实验组。两组细胞经24h培养后,运用免疫组化染色法(SP法)观察TSP-1、TGF-β1及EGFR在肺动脉内皮细胞中的表达情况。结果低氧刺激24h后的内皮细胞TSP-1主要表达部位在胞浆内,阳性表达率为80.00%,与常氧对照组的20.00%阳性率相比差异有统计学意义(P<0.01)。TGF-β1阳性部位在胞核和胞浆中,低氧实验组和常氧对照组阳性表达率分别为92.50%和25.00%,组间差异有统计学意义(P<0.01)。EGFR主要表达部位在胞膜,低氧实验组和常氧对照组阳性表达率分别为89.74%和15.00%,组间差异有统计学意义(P<0.01)。结论低氧刺激后肺动脉内皮细胞高表达TSP-1、TGF-β1、EGFR,上述因子表达增加可能与低氧性肺动脉高压有关。
Objective To investigate the expression of thrombospondin- 1,transforming growth factor- β 1 and epidermal growth factor receptor in the hypoxia pulmonary arterial endothelial cells. Methods The pulmonary arterial endothelial cells from SD rat were primarily cultured. The subculturing cells were divided into two group,the normoxia cells as control group and the hypoxia cells( 3% oxygen concentration) as experimental group. The expression of TSP- 1,TGF- β 1 and EGFR were measured by immunohistochemical staining method in the pulmonary arterial endothelial cells after 24 hours cultured. Results The TSP- 1 positive expression site of pulmonary arterial endothelial cells was in the cytoplasm and the positive rate was 80. 00%, which was significantly stronger than those of controls( 20%)( P < 0. 01). The expression of TGF- β 1 was in the nucleus and cytoplasm and the positive rate was 92. 50%,which was significantly stronger than those of controls( 25%)( P < 0. 01). The EGFR positive expression site is near the membrane and EGFR- positive expression rate was 89. 74%,which were significantly stronger than those of controls( 15%)( P < 0. 01). Conclusion The expression of TSP- 1,TGF- β 1 and EGFR appears to be increased in hypoxic conditions, which may contribute to the pathogenesis of hypoxic pulmonary vascular remodeling.
