摘要
目的建立一种灵敏、快速的反相高效液相色谱电化学检测方法(RP-HPLC-ECD),测定红葡萄酒中的顺式和反式白藜芦醇.方法色谱条件经过优化,用C18柱(250×4.6mm,5μm)作为分析柱,流动相为20mM柠檬酸钠(pH4.0):甲醇(50:50),柱温为35℃,流速为1ml/min,电化学检测器工作电压为0.75V.结果最低检测限为5 ng/mL,线性范围为10~50000 ng/mL,顺式和反式白藜芦醇重复5次进样的相对标准差(RSD)分别为3.52%和3.16%.结论该方法简便、快速、重复性好,适用于食品及生物样品中白藜芦醇含量的分析.
Object:A sensitive and rapid high-performance liquid chromatography with electrochemistry detection method was developed for the determination of resveratrol in red wine.Method:Chromatographic separation was achieved using a C18column(250×4.6mm,5μm)with a mobile phase containing20mM sodi-um citrate(pH4.0)and methanol(50∶50)at a flow rate of1.0mL/min.The voltage of detection was set at0.75V.Results:The limit of detection was5ng/mL at a signal-to-noise ratio of5:1and the limit of quan-tification was10ng/mL.The linearity of calibration curve was from10ng/mL to5000ng/mL.The relative standard deviations(RSD)of5replicates were3.52%and3.16%for cis-resveratrol and trans-resveratrol,re-spectively.Conclusion:This method is simple,rapid and reproducible and suitable to the determination of resveratrol in food and biological samples.
