摘要
目的探讨丹参素和丹酚酸B对大鼠骨髓基质干细胞(MSCs)成骨与成脂分化的影响。方法分离纯化大鼠MSCs,观察不同浓度丹参素和丹酚酸B对成脂分化率、脂蛋白脂酶mRNA表达和碱性磷酸酶(ALP)活性影响。结果丹参素和丹酚酸B处理明显减少MSCs中脂肪细胞数、油红染色比色值和脂蛋白脂酶表达,但增加ALP活性(P<0.01)。结论丹参素和丹酚酸B均能抑制大鼠MSCs成脂分化,并促进成骨分化。
Objective To investigate the effect of tanshinol and salvianolic acid B(SalB) on osteogenesis and adipogenesis of rat marrow stroma stem cells(MSCs). Methods Rat MSCs were isolated and purified. The effect of different concentrations of tanshinol and SalB on adipogenic differentiation, lipoprotein lipase mRNA and alkaline phosphatase(ALP). Results Tanshinol and SalB treatment decreased the adipocyte number, Oil Red O absorbance and lipoprotein lipase mRNA expression but increased ALP activity in MSCs compared with control groups(P<0.01). Conclusion Tanshinol and SalB can inhibit the adipogenesis and promote the osteogenesis in rat MSCs.
