摘要
建立海带药材的HPLC参考指纹图谱。运用DAD-Agilent 1100高效液相色谱仪,色谱条件:Agilent Zorbax XDB C18柱(4.6 mm×250 mm;5.0μm)色谱柱,1.0 mL.min-1流速,230 nm检测波长,360 nm参比波长,20μL进样量,25℃柱温,乙腈(A)-0.1%磷酸水(B)梯度洗脱。结果显示,建立的指纹图谱特征明显,化学信息完整,包含11个共有峰。方法的系统适应性测定结果符合指纹图谱的技术规范。研究表明,首次建立的海带HPLC参考指纹图谱可用于海带药材的真伪鉴别。
Objective to establish reference fingerprint of Laminaria japonica.Methods the reference fingerprint were determined by Agilent 1100 DAD-HPLC on Zorbax XDB C18 column(250 mm×4.6 mm,5 μm),with gradient elution in methanol-water(0.7%H3PO4),column temperature was at 30 ℃,flow rate was 1.0 mL/min,wavelength was at 280 nm,and the inject volume was 20 μL.Results the reference fingerprint of L.japonica,which included 11 common peaks and revealed good fingerprint characteristic,was built by high performance liquid chromatography for the first time.The tests of precision, stability and repeatbility for the method was according with the fingerprint technique criterion.Conclusion the reference fingerprint could be used to identification of L.japonica.
